摘  要:目的探讨槟榔碱诱导上皮细胞HaCaT基质金属蛋白酶9(MMP9)表达的分子机制。方法槟榔碱处理上皮细胞HaCaT,Western blot检测MMP9、核因子(NF)-KB、激活蛋白(AP)-1和Ets-1蛋白的表达,RNAi实验初步确定槟榔碱诱导下调控MMP9表达的转录因子。结果槟榔碱呈时间和剂量依赖性上调上皮细胞HaCaT MMP9的表达,并促进转录因子NF-κB、AP-1和Ets-1的表达。siRNA下调NF-κB表达后,MMP9的表达明显受到抑制,而siRNA下调Ets-1和AP-1的表达后,MMP9的表达下调但不明显。结论槟榔碱通过上调上皮细胞HaCaT NF-κB、AP-1和Ets-1等转录因子的表达促进MMP9的表达上调。初步确定NF-κB是槟榔碱诱导下促进MMP9表达上调的主要转录因子。Objective This study was designed to investigate the mechanism of arecoline-induced matrix metalloproteinase 9(MMP9) expression in epithelial HaCaT cells. Methods HaCaT cells were treated with arecoline having different concentrations at different times. MMP9, nuclear factor(NF)-κB, activator protein(AP)-1, and Ets-1 protein expressions were detected by western blot analysis. Transcription factors of regulated MMP-9 expression induced by arecoline were detected by RNA interference test. Results Arecoline upregulated MMP9 expression in HaCatcells through the modes of dosage and time symbiosis. Furthermore, arecoline upregulated NF-κB, AP-1, and Ets-1 protein expressions. As detectedby the siRNA test, arecoline affected MMP9 through transcription factors, such as NF-κB, AP-1, and Ets-1, especially NF-κB..Conclusion Arecoline can upregulate MMP9 expression by promoting NF-κB, AP-1, and Ets-1 expressions.and primarily through NF-κB transcription.
