低分子量肝素对人肺腺癌A549细胞作用的影响研究

摘　　要：目的观察低分子肝癌(LMWH)对人肺腺素A549细胞体外作用的影响,阐明LMWH抑制肿瘤血管生成的可能机制。方法采用逆转录-聚合酶链反应(RT-PCR)半定量检测LMWH处理细胞后血管内皮生长因子(VEGF)mRNA的表达变化。结果 VEGF mRNA水平的表达分别是随药物浓度增加和时间的延长而逐渐减弱,差异具有显著性(P<0.05)。结论 LMWH可以下调细胞VEGF mRNA的表达水平,可作为血管生成抑制剂有望成为解决肿瘤细胞耐药和肿瘤靶向治疗的新途径。Objective To study the effects of low molecular weight heparin（ LMWH） on angiogenesis of human lung cancer cell line A549 and its mechanisms. Methods A549 cells were incubated in culture medium in vitro. The expression of vascular endothelium growth factor（ VEGF） gene was analyzed by RT-PCR after being treated with LMWH. Results Compared to the control group,the expression VEGF mRNA was decreased markedly in the A549 cells exposed to LMWH（ P〈0. 05）. Conclusion LMWH may inhibit the angiogenesis abilities of A549 cells by down-regulating the expression of VEGF mRNA.
