大株红景天注射液对大鼠心肌缺血预处理血管内皮生长因子的影响

摘　　要：目的研究大株红景天注射液对大鼠急性心肌缺血预处理后血管内皮生长因子(VEGF)表达的作用机制。方法大鼠24只随机分为对照组、模型组、缺血预处理组。模型组、缺血预处理组经尾静脉注射垂体后叶素制作大鼠急性心肌缺血模型,分别于造模前、造模1 h及治疗1周后采用双抗体夹心ELISA法测定各组大鼠胎肝激酶1(FLK-1)、缺氧诱导因子-1α(HIF-1α)、VEGF基因及蛋白表达情况。结果缺血预处理组在造模1 h及治疗1周后FLK-1及FLK-1mRNA、HIF-1α及HIF-1αmRNA、VEGF及VEGF mRNA的表达明显增高,与对照组、模型组及同组缺血预处理前比较差异均有统计学意义(P<0.05)。结论大株红景天注射液可促使急性心肌缺血大鼠缺血心肌血管内皮新生,减轻血管内皮炎症反应及损伤程度,对大鼠急性心肌缺血再灌注损伤有一定的保护作用。Objective It is to study the effect of Rhodiola rosea injection on the expression of vascular endothelial growth factor （VEGF） in rats undergoing acute myocardial ischemia preconditioning and its mechanism. Methods 24 rats were ran- domly divided into control group, model group, ischemie preconditioning group. Rat models of acute myocardial ischemia were established by tail vein injection of pituitrin in model group and isehemie preconditioning group. The expression of fetal liver kinase 1 （ FLK - 1 ） , hypoxia inducible factor -1α （ HIF -1α） , VEGF protein and mRNA, were determined by double anti- body sandwich EUSA method in every group before model establishing, lh after establishing and 1 week after treatment. Re- suits The expression of FLK - 1 and FLK - 1 mRNA, HIF - 1α and HIF -lct mRNA, VEGF protein and mRNA increased significantly in ischemie preconditioning group in lh after establishing and l week after treatment, and compared with that in control group, model group and before ischemic preconditioning, the differences were significant （P 〈 0.05 ）. Conclusion Rhodiola erenulate injection can stimulate the growth of neovascular endothelial cells, and relieve the vascular endothelial in- flammation and injury degree in the rats with acute myocardial ischemia, so it has some protecting effect on rat myocardial is- chemia reperfusion injury.
