脱蛋白对异种骨中骨形成蛋白影响的实验研究

摘　　要：目的研究脱蛋白对异种骨中骨形成蛋白的影响。方法取猪骨制成骨块脱脂后,用双氧水和胰蛋白酶脱蛋白,免疫组化检测骨中骨形成蛋白表达情况。结果双氧水脱蛋白后,骨中的骨形成蛋白表达较低,而胰蛋白酶脱蛋白后骨形成蛋白表达与新鲜骨类似。结论使用酶法脱蛋白既能有效的去除骨中具有抗原性的胶原蛋白成分,又能保留骨中的骨诱导活性物质。Objective To study the influence of deprotein on bone morphogenetic protein （BMP） in xenobone. Methods After defatting porcine bone, we deproteined the porcine bone with hydrogen peroxide and trypsin. Then the BMP expression was measured with immunohistochemistry. Results After depro/einization with hydrogen peroxide, the BMP expressed less than deproteinization with trypsin, and it is similar with fresh bone after deproteinization with trypsin. Conclusion Deproteinization with enzyme is a good way to remove antigen of protein and keep the osteoinductive ingredients.
