原花青素减轻大鼠蛛网膜下腔出血后早期脑血管痉挛的实验研究

摘  要:目的探讨原花青素对大鼠蛛网膜下腔出血(subarachnoid hemorrhage,SAH)后早期脑血管痉挛(cerebral vasospasm,CVS)的影响及其作用机制。方法雄性SD大鼠60只,随机分为假手术组、SAH组、生理盐水治疗组和原花青素治疗组,每组15只。采用后颅窝注血法制作大鼠SAH模型,原花青素治疗组予原花青素100 mg/kg腹腔注射,q12h,生理盐水治疗组注射等量生理盐水。造模后24h行神经行为学评分,取脑干组织行HE染色后测量基底动脉直径及壁厚,化学比色法测定诱导型一氧化氮合成酶(inducible nitric oxide synthase,iNOS)水平,免疫组化法检测脑干内皮素-1(endothelin-1,ET-1)表达。结果与SAH组比较,原花青素治疗组大鼠神经行为学评分增高,基底动脉直径增大,管壁变薄,iNOS和ET-1表达减少,差异均有统计学意义。结论原花青素能减轻大鼠SAH后早期CVS,其机制可能与抑制SAH后iNOS和ET-1的过度表达有关。Objective To assess the effect of procyanidin on early cerebral vasospasm after subarachnoid hemorrhage in rats.Methods 60 healthy SD rats were randomly divided into 4groups:sham-operated,SAH without treatment,SAH treated with vehicle(NS),SAH treated with procyanidin(100mg/kg,q12h).SAH model was produced by injecting autologous blood into the posterior cranial fossa of rats.24 hafter SAH induction the neurologic scores were recorded and then the rats were sacrificed.Brain stems were removed and HE stained for measurement of luminal diameter and wall thickness of the basilar arteries.The expression of iNOS was determined by colorimetry.The expression of ET-1was determined by immunohistochemistry.Results Compared with SAH group,the neurologic scores of rats treated with procyanidin were significantly higher.The basilar arteries of rats treated with procyanidin were significantly thinner and the luminal diameters significantly longer.The expression of iNOS and ET-1 were significantly reduced.Conclusion Procyanidin attenuates early cerebral vasospasm and improves the neurological deficits after subarachnoid hemorrhage in rats by inhibiting the expression of iNOS and ET-1.
