脐血来源的细胞因子诱导的杀伤细胞对人肾癌ACHN细胞系体外杀伤效果的实验研究

摘　　要：目的探讨脐血来源的细胞因子诱导的杀伤细胞（CIK）体外对人肾癌ACHN细胞系的体外杀伤作用。方法采集脐血单个核细胞，获得纯度〉90％的脐血单个核细胞（CBMNC），细胞数量需达到2×10^8个；加入干扰素γ，CD3单克隆抗体和白细胞介素2刺激CBMNC增殖成为CIK细胞；流式细胞仪检测CIK细胞表面CD3，CD56等分子的表达并进行质检，最终用乳酸脱氢酶试剂盒检测其对ACHN细胞的杀伤率。结果脐血CIK细胞对ACHN细胞的杀伤率在效靶比为1：5、1：10及1：20时可分别达到（22．2±4．0）％、（41．2±3．7）％、（59．8±2．4）％。结论脐血CIK细胞对人肾癌细胞有杀伤效果，并且随着效靶比的增加而增强，对临床使用脐血CIK细胞治疗肾癌有一定指导意义。Objective To investigate the anti-tumor effects of cytokine-induced kill cells （CIK） against human renal carcinoma cell line ACHN in vitro. Methods Monocytes from cord blood were collected to acquire high purity of the cord blood mononuclear cells （CBMC）. The cell surface markers CD3 and CD56 were detected by flow eytometer. Lactate dehydrogenase kit was used for detect the execution ratio to ACHN cell line. Results Umbilical cord blood CIK cells on ACHN cells killing rate was achieved （22.2 ±4.0）% , （41.2 ±3.7）% and （59.8 ±2.4） % when the effect/target ratio was 1 ： 5, 1 ： 10 and 1 ： 20. Conclusion CIK cells of the cord blood have strong anti-cancer activity against ACHN ceils in vitro.
