摘 要:目的旨在研究CTX联合BUS对Wister小鼠卵巢及早期肺功能损伤组织结构的影响。方法选取2-3月龄的雌性Wister小鼠60只,随机分成四组:A正常对照组、B生理盐水组、C组CTX+BUS组、D组CTX+BUS+生理盐水组。C、D两组第1天以腹腔注射CTX负荷剂量50 mg/(kg·d),第2天开始8 mg/(kg·d)连续注射14 d,后皮下注射BUS10 mg/(kg·w),共2次。C组小鼠于第15天处死。D组于第15天开始注射生理盐水8 mg/(kg·d),共15 d,于第30天处死余组小鼠。在处理期间观察小鼠的一般情况、体重、各级卵泡数水平。以化学发光法测定各组小鼠血清中E2及FSH的水平,观察卵巢及肺组织常规病理切片下的形态学变化。结果与A、B组卵巢相比,C组E2显著下降、FSH水平显著增加、各级卵泡数量减少(P〈0.05)。C、D组相比,E2下降、FSH增加(P〈0.05),两组各级卵泡计数虽减少,但差异无统计学意义。A、B组中E2、FSH、卵泡数量比较,差异无统计学意义。C组与A、B、D组肺组织对比,肺组织血管内皮细胞水肿,肺泡间隔内可见渗出液。A、B、C组肺组织形态无异常。结论 CTX+BUS使原始卵泡和初级卵泡的显著减少可能与小鼠卵巢中血E2下降有关,并且CTX+BUS也会造成一过性肺损伤。Objective To assess the effects of cyclophosphamide(CTX) and busulfan(BUS) on ovarian injuries and initial injury of lung. Methods Sixty female SD rats age of 2 to 3 months were divided into four groups: group A(control group), group B(saline group), group C(CTX+ BUS group), group D(CTX+BUS+saline group). SD rats of group C and group D were processed by intraperitoneal injection of CTX 50 mg/(kg·d) at the 1st day and then 8 mg/(kg·d) from day2, for 14 days. SD rats of group C and group D were processed by subcutaneous injection of BUS 10 mg/(kg·w) for twice, and SD rats of group C were killed at day 15, whereas SD rats of group D had been processing by 8 mg/(kg·d)saline for another 15 days, and all of SD rats were killed at day 30. General conditions, weight, number of levels of folliole of rats of four group were observel every day. Estrogen(E2) and follicular stimulating hormone(FSH) were detected by chemiluminescence method. The lung and ovaries of each group were prepared for HE sections. Results Compared with A, B group of ovarian, levels of E2, number of levels of follicular, FSH in the group C decreased and increased significantly(P〈0.05). Compared with group D, C, E2 decreased, FSH increased(P〈0.05), two groups of follicle count at all levels was reduced, but there was no statistically significant difference. In group B and A, E2, FSH, number of follicles were compared, there was no statistically significant difference. Group C compared with group A, B, D in lung tissue, lung tissue vascular endothelial edema, can be seen in the alveolar interval exudate. Lung tissue form of A, B, C group without exception. Conclusion CTX together with BUS can induce primodial follicles and primary follicles undergo atresia, which can be close to the decreased level of E2. CTX together with BUS can also induce transitory pulmonary injury in early stage.
