摘　　要：目的探讨Aurora A激酶抑制剂VX-680对人宫颈癌细胞化疗敏感性的影响。方法以Aurora A激酶抑制剂VX-680抑制宫颈癌细胞株Caski中Aurora A的表达,再将宫颈癌细胞用不同浓度顺铂处理,Western Blot检测Aurora A蛋白受抑制情况,用流式细胞仪分析Aurora A表达下调后宫颈癌细胞生长抑制率和细胞凋亡率的变化。结果 Aurora A表达下调使顺铂对宫颈癌细胞的生长抑制率和细胞凋亡率均升高(P均<0.01)。结论 Aurora A激酶抑制剂VX-680可使人宫颈癌细胞对顺铂的敏感性增强,抑制Aurora A可望提高顺铂对宫颈癌的治疗效果。Objective To explore the effect of Aurora A inhibitor VX-680 on the treatment efficacy of Cisplatin for cervical cancer.Methods Aurora A inhibitor VX-680 was used to down regulated Aurora A in cervical cancer cell line Caski.After treated with different dose of Cisplatin,Aurora A expressions were detecfed by western Blot,cell growth and apoptosis were detected by flow cytometry.Results Down-regulation of Aurora A expression by VX-680 increased the cell inhibition and apoptosis when cells were treated with Cisplatin.Conclusion There is a positive relationship between Aurora A expression and chemosensitivity in cervical cancer cell,suggesting that inhibiting Aurora A expression may enhance the treatment efficacy of Cisplatin for cervical cancer.
