摘 要:目的研究正加速度(positive acceleration,+Gz)暴露对实验性胃溃疡大鼠胃黏膜降钙素基因相关肽mRNA(calcitonin gene-related peptide-mRNA,CGRP-mRNA)的表达及胃黏膜血流(gastric mucosal blood flow,GMBF)的影响。方法选择雄性SD大鼠40只随机分成4组,每组10只,A组为正常对照组,不做处理;对B、C、D组大鼠采用乙酸烧灼法建立胃溃疡模型,造模3 d后加速度处理大鼠,B组地面捆绑5 min(相当于+1 Gz恒速暴露),C组+5 Gz旋转5 min,D组+10 Gz旋转5 min,隔日1次,共4次。1周后麻醉大鼠并固定,用MoorVMS-LDF2型激光多谱勒血流仪测定GMBF;采集胃黏膜组织,用荧光定量PCR的方法定量测定胃黏膜CGRP-mRNA含量(2^-△△ct)。结果胃黏膜CGRP-mRNA的含量,B组(3.090±0.154)较A组(0.966±0.089)升高(P<0.05);+Gz暴露后,B组>C组(2.262±0.116)>D组(1.096±0.077),差异均有统计学意义(P<0.05)。GMBF量,A组(432.919±11.598)pU>B组(367.602±10.133)pu>C组(303.925±16.417)pu>D组(246.750±22.815)pu,差异均有统计学意义(P<0.05)。胃溃疡大鼠(B组、C组、D组大鼠)+Gz暴露后,CGRP-mRNA表达和GMBF量存在正相关(r=0.807,P<0.05)。结论 +Gz暴露导致实验性大鼠胃溃疡愈延迟合,其可能与+Gz条件下胃黏膜血流量减少及胃黏膜CGRP-mRNA的表达改变有关。Objective To study the effects of positive acceleration(+Gz) on CGRP-mRNA in gastric mucosal and gastric mucosal blood flow of rats with experimental gastric ulcer and the related mechanism of the effect on ulcer healing.Methods Forty male SD rats were randomly and equally divided into four groups.Rats in group A was the normal control group without exposure to constant+Gz.The gastric ulcer was induced with acetic acid in rats of group B,C,D.After 3 d,rats in group B were bound on the ground for5min(exposure of constant +1 Gz).Accordingly,the rats of group C and D were respectively exposed to + 5 Gz and + 10 Gz for 5min(once every other day,4 times in all).After 1 week,GMBF of rats which were anesthetized and fixed firstly was measured with the MoorVMS-LDF2 laser doppler blood flow meter.Gastric mucosa were collected to determinate quantitatively CGRP-mRNA in gastric mucosal by fluorescence quantitative PCR.Results The content of CGRP-mRNA of rats in group B(3.090±0.154) was higher than that in group A(0.966 ± 0.089,P 0.05).After exposure to + Gz,the content of CGRP-mRNA of rats in group B group C(2.262±0.116) group D(1.096±0.077),and all differences were statistically significant(P 0.05).For the content of GMBF,group A(432.919± 11.598) pUgroup B(367.602±10.133) pU group C(303.925± 16.417) pU group D(246.750 ± 22.815) pU.The differences were statistically significant(P 0.05).The content of CGRP-mRNA and GMBF in gastric mucosal of rats(rats in group B,C,D) with gastric ulcer had positive correlation after exposure to different +Gz(r=0.807,P 0.05).Conclusion + Gz exposure delays gastric ulcer healing of rats,which may be related to the reduction of GMBF and alteration in the expression of CGRP—mRNA after exposure to+Gz.
