摘  要:目的探讨槲皮素对刀豆蛋白A(Concanavalin A,Con A)诱导小鼠急性肝损伤的保护作用及其可能的分子机制。方法通过检测Con A注射后8 h血清转氨酶水平及肝组织病理学评估肝损伤;应用酶联免疫法(ELISA)检测TNF-α、IFN-γ、IL-4水平;定量PCR和Western blotting或免疫组织化学检测肝组织mRNA与蛋白的表达。结果与模型组相比,槲皮素预防组显著降低了血清转氨酶水平(P<0.05),且组织病理学分析表明肝脏炎症坏死程度明显减轻;与模型组相比,槲皮素预防组显著降低了血清促炎症细胞因子TNF-α、IFN-γ和IL-4的水平(P<0.01);槲皮素预防组肝组织NOX4、COX-2和i NOS基因与蛋白的表达均明显低于模型组(P<0.05)。结论槲皮素对Con A诱导小鼠急性肝损伤具有保护作用,其发挥有益作用的机制可能与抑制肝脏的炎症反应有关。Objective To investigate the hepatoprotective effect and the potential molecular mechanisms of quercetin on Concanavalin A ( ConA)-induced acute liver injury in mice. Methods The liver injury was examined by measuring serum transaminase 8 h after ConA injection and liver histology. The tumor necrosis factor-α ( TNF-α) , interferon-γ( IFN-γ) , and interleukin-4 ( IL-4 ) levels in liver tissue were detected by enzyme linked immunosorbent assay (ELISA). Furthermore, the gene and protein expression of NADPH oxidase 4 (NOX4), cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase ( iNOS) in liver were detected by Real-time quantitative PCR and Western blotting or immunohistochemical staining, respectively. Results Quercetin significantly decreased the elevated plasma levels of aminotransferases compared with model group ( P〈0 . 05 ) . Histopathological analysis also showed that quercetin preven-ted inflammation and necrosis. Also, quercetin pretreatment reduced the serum levels of pro-inflammatory molecules such as TNF-α, IFN-γ and IL-4 compared with the vehicle controls (P〈0. 05). Furthermore, the expression levels of NOX4, COX-2 and iNOS mRNA or protein in liver tissues were significantly lowered by quercetin pretreatment. Conclu-sion Quercetin pretreatment protects against ConA-induced acute liver injury in mice, and the beneficial effect of quer-cetin may be related to inhibition of the inflammatory response in liver.
