摘 要:目的观察趋化因子CXCL12的特异性受体CXCR7及核转录因子-κB(nuclear factor kappa B,NF-κB)在乳腺癌组织中的表达情况,探讨其与各临床病理指标的意义,研究其与乳腺癌的发生及淋巴结转移的关系。方法收集甘肃省人民医院2003-01-01-2006-01-01手术切除的102例浸润性乳腺癌标本,以癌旁正常组织作为对照组,应用免疫组织化学及荧光定量PCR的方法,检测CXCR7及NF-κB在乳腺癌组织及癌旁正常组织中的表达水平的差异;应用χ2检验比较乳腺癌组织及癌旁正常组织中CXCR7及NF-κB的表达差异,Spearman秩相关分析CXCR7及NF-κB的表达与各临床病理指标之间的相关性。结果荧光定量PCR结果显示,乳腺癌组织CXCR7mRNA的表达水平明显高于癌旁正常组织,P=0.017。免疫组化结果显示,CXCR7在乳腺癌和癌旁正常组织中的表达水平分别为86.3%(88/102)和14.7%(14/102),差异有统计学意义,χ2=28.00,P<0.001;NF-κB在乳腺癌和癌旁正常组织中的表达水平分别为89.2%(91/102)和10.8%(11/102),差异有统计学意义,χ2=22.00,P<0.001。CXCR7和NF-κB的表达水平与患者年龄、分化程度和肿瘤大小等无关,与淋巴结转移有关,P值分别为0.023和0.003;CXCR7及NF-κB的表达水平呈正相关性,r=0.628,P=0.048。结论 CXCR7和NF-κB在乳腺癌组织中具有相关性高表达,在乳腺癌的发生中可能起着重要作用,且与淋巴结的转移存在密切相关,对评估预后具有重要意义。OBJECTIVE To investigate the expression and relationship of CXCR7 and nuclear factor kappa B (NF-κB) in breast carcinoma and the prognosis.METHODS RT-PCR and IHC were used to detect the expressions and relationship of CXCR7 and NF-κB in 102 samples of human breast carcinoma from Gansu Province People's Hospital to analyze the value of CXCR7 and NF-κB in predicting the prognosis and metastasis of patients.RESULTS Immunofluorescence (IF) analysis revealed that the expression level of CXCR7 and NF-κB were found up-regulated in breast carcinoma tissues as compared with normal tissues (P=0.017).The expression of CXCR7 was 86.3% (88/102) and 14.7% (14/102) in carcinoma and normal tissues by immunohistochemical technique(x2 =28.00,P<0.001).The expression of NF-κB was 89.2%(91/102) and 10.8%(11/102).They were significantly different (x2 =22.00,P<0.001).They were related to lymph node metastasis (P=0.023,P=0.003),other than the age,differentiation and large of tumer.The expression level of CXCR7 was related to the NF-κB (r=0.628,P=0.048).CONCLUSIONS Overexpressions of CXCR7 and NF-κB are correlated with the biologic behavior and prognosis of breast carcinoma.The CXCR7 and NF-κB may play important roles in lymph node metastasis of breast cancer.Determination of CXCR7 and NF-κB is helpful in predicting of metastasis the prognosis of breast carcinoma.
