摘  要:目的:制备实验性自身免疫性肌炎(EAM)动物模型,观察白介素-6(IL-6)在EAM模型肌肉组织中的表达。方法:豚鼠20只,随机分为模型组12只和对照组8只,模型组采用纯化兔骨骼肌肌球蛋白+弗氏完全佐剂(CFA)免疫注射,对照组给予CFA+PBS免疫注射,2组均每周免疫1次,连续6周,并同时给予2次百日咳菌液腹腔注射。观察2组临床表现、血清肌酶水平及肌肉病理,并采用免疫组织化学方法检测IL-6在肌肉组织中的表达。结果:模型组豚鼠血清中肌酸激酶、乳酸脱氢酶、谷草转氨酶水平均高于对照组,差异有统计学意义(P<0.05)。注射免疫第6周末,2组临床评分差异有统计学意义(P<0.05),模型组血清肌酶水平与临床评分呈正相关(r=0.853,P<0.05)。模型组的肌束膜、肌内膜和坏死肌纤维处均有IL-6阳性表达,对照组肌肉标本几乎无IL-6表达。结论:EAM模型制备成功,IL-6在其发病过程中可能起重要作用。Objective:To establish an animal model of experimental autoimmune myositis(EAM), and to observe the expression of IL-6 in the muscle of animals. Methods: Twenty guinea-pigs were randomly assigned into EAM group(n=12) and control group(n=8). The EAM group was subcutaneous injected with rabbit skeletal muscle myosin plus Freund's complete adjuvant(CFA), and the control group was received CFA and equal amount of PBS.Each guinea-pigs received twice injection of pertussis toxin.The animals in both groups were immunized once a week, lasting for 6 weeks. The clinical manifestation, the serum muscular enzymes levels and pathology of the skeletal muscles were observed. The immunohistochemistry technique was used to detect the expression of IL-6 in the muscle tissue. Results: The serum levels of creatine kinase, lactate dehydrogenase and aspartate aminotransferase of the EAM group were increased significantly, compared with those of the control group(P〈0.05). At the6 th week, there was statistical difference of the 2 groups in the clinical scores, and the serum muscular enzyme levels were positively correlated with the clinical scores in the EAM model group(r=0.853, P〈0.05). In the perimysium, endomysium, necrotic fibers of the EAM group, the level of IL-6 was dramatically up-regulated compared with control group with rare expression. Conclusion: The establishment of EAM animal model is successful. IL-6may play an important role in the pathogenesis of EAM.
