小干扰RNA沉默β-catenin基因对人结肠癌SW480细胞增殖及凋亡的影响

摘　　要：目的:探讨小干扰RNA(siRNA)沉默人结肠癌SW480细胞β-catenin基因表达对SW480细胞增殖和凋亡的影响。方法:采用脂质体转染法合成靶向β-catenin的双链siRNA(β-catenin-siRNA),转染SW480细胞,RTPCR及Western blotting检测SW480细胞中β-catenin mRNA及蛋白的表达,MTT实验和流式细胞术分别检测SW480细胞的增殖和凋亡。结果:RT-PCR及Western blotting检测结果显示β-catenin-siRNA转染后,与空白组、阴性对照组及实验组相比,β-catenin-siRNA转染组SW480细胞中β-catenin mRNA水平明显下降,其蛋白表达也明显下调。β-catenin-siRNA转染后,SW480细胞的凋亡率明显高于阴性对照组及实验组,细胞增殖能力明显下降。结论:siRNA沉默β-catenin的表达可诱导SW480细胞凋亡,抑制细胞的增殖,为结肠癌的临床治疗提出新的思路,可以作为结肠癌治疗的一个新靶点。Objective：To investigate the effects of small interfering RNA（siRNA）targeting theβ-catenin gene in wnt/β-catenin signaling pathway on the apoptosis,proliferation and invasion of human colon cancer cell line SW480.Methods：Synthesis of targeted double stranded siRNAβ-catenin（β-catenin-siRNA）was transfected into SW480 cells.The expression ofβ-catenin mRNA and protein in SW480 cells were detected by RT-PCR and Western blotting.The proliferation of SW480 cells was analyzed by MTT assay,and the cell apoptosis was analyzed by flow cytometry.The caspase-3detective kit and invasion assay were performed to detect the change of caspase-3activity and invasion in SW480 cells.Results：After transfection ofβ-catenin-siRNA,there was a significant decrease in the expressions of bothβ-catenin mRNA andβ-catenin protein in theβ-catenin-siRNA group,as compared with the blank and negative control and liposome groups.The growth of SW480 cells in theβ-catenin-siRNA group was significantly lower than that in the blank and negative control and liposome groups,and the rate of apoptosis was obviously increased.The activity of caspase-3in theβ-catenin-siRNA group was decreased compared with thatin the blank and negative control and liposome groups.Conclusion：The siRNA silencingβ-catenin expression in SW480 cells can suppress the proliferation,and induce the apoptosis.
