摘 要:目的 考察宣木瓜醇提物活性成分在大孔树脂不同极性洗脱液中分布,为系统分离宣木瓜有效部位提供实验资料和科学依据。方法 采用AB-8大孔树脂和不同浓度乙醇液洗脱分离,显色或沉淀反应检识不同极性洗脱液中的化学成分,分光光度法测定总多酚、总多糖含量,高效液相色谱法测定绿原酸和原儿茶酸以及齐墩果酸和熊果酸含量。结果 总多糖主要分布在水洗脱液中;总多酚、绿原酸和原儿茶酸主要分布在30%~50%乙醇洗脱液中;齐墩果酸和熊果酸主要分布在95%乙醇洗脱液中。结论 采用的分离方法步骤简单,具有很好的重现性,实验结果为宣木瓜醇提物的进一步研究奠定了基础。Objective To investigate the distribution of active ingredients in ethanol extracts of Chaenomeles Fructus in the macroporous resin elution of different polarity,and provide the experimental data and scientific basis for the systematic separation of the effective part of Chaenomeles Fructus. Methods Using AB-8 macroporous resin and different concentrations of ethanol solution to separate,chemical components in eluents of different polarity were demonstrated though precipitation reaction and chromogenic reaction. The content of total polysaccharides,total polyphenols and protocatechuic acid,chlorogenic acid,oleanolic acid,ursolic acid were determined by spectrophotometry and high-performance liquid chromatography,respectively. Results TPS is mainly distributed in the water eluent; TPP,CA and PA were in the 30%- 50% ethanol eluent; OA and UA were in 95% ethanol eluent. Conclusion The separation step of this experiment is uncomplicated and has good reproducibility,and it makes a foundation for further research and development of ethanol extracts of Chaenomeles Fructus.
