摘  要:为研究不同宿主来源的新城疫病毒(NDV)的生物特性及分子特征,本研究比较了两株(Ch/CH/SD/2008/128和Du/CH/SD/2009/134)同属ClassⅡ基因VII型的NDV分离株的生物学特性,并采用RT-PCR方法扩增及测定两株病毒的全基因组序列。结果显示,两株病毒对雏鸡具有相同的高致病性,但鸡源分离株Ch/CH/SD/2008/128对雏鸭致死率高达70%,而鸭源分离株Du/CH/SD/2009/134对雏鸭致死率仅为20%。全基因组序列比较分析显示:两株病毒的基因组长度均为15 192 nt,裂解位点为112RRQKQF117,属于典型的强毒特征,并且两者之间同源性超过96%。Ch/CH/SD/2008/128的F和HN蛋白中某些氨基酸位点与其他VII型病毒存在差异,这可能是导致该病毒对鸭具有高致病性的原因。结果表明两株不同宿主来源的ClassⅡ基因VII型NDV在遗传信息方面差异不大,但对鸭的致病力具有明显差异。To illustrate the relationship between viral pathogenesis and host specificity among Newcastle disease virus (NDV) isolates from different hosts, the genomic characterization and the pathogenicity of 2 NDV isolated from chicken (Ch/CH/SD/ 2008/128) and duck (Du/CH/SD/2009/134) were compared in this study. The results of genomic scquceing showed that both isolates were consist of 15,192 nucleotides, and the deduced amino aicd sequence analysis indicated that the 2 virus had the same virulent motif U2RRQKQF^117 at the F protein cleavage site. The phylogenetic analysis revealed that both isolates were clustered into Class lI NDVs. In addition, they were both highly pathogenic to chickens, while, the morality of ducks infected by Ch/CH/SD/ 2008/128 and Du/CH/SD/2009/134 was 70% and 20%, respectively. The data indicated that the isolates from different hosts share the same genotype with a few divergences in the genetic information, but their pathogenicity for ducks were significant different.
