TRIM28对肺鳞癌SK-MES-1细胞生长的影响

摘　　要：目的:探讨TRIM28基因对肺鳞癌细胞系SK-MES-1细胞生长的影响。方法:采用RNA干扰技术抑制肺鳞癌SK-MES-1细胞TRIM28基因的表达,通过RT-PCR和Western-blot技术检测抑制效果,应用克隆形成实验和MTT实验观察TRIM28基因被抑制后SK-MES-1细胞的增殖和活性。结果:RNA干扰技术能有效抑制TRIM28基因在肺鳞癌SK-MES-1细胞中的表达。TRIM28基因被抑制后,肺鳞癌SK-MES-1细胞的活性和生长受了到明显抑制。结论:TRIM28对肺鳞癌SK-MES-1细胞的生长具有促进作用。Objective：To study the influences of TRIM28 gene on growth of lung squamous cell carcinoma SKMES-1 cells.Methods：TRIM28 gene was inhibited by RNA interference assay, RT-PCR and Western-blot was used to detect the inhibitory effects. After the TRIM28 gene was inhibited, the proliferation and activity of SK-MES-1 cells were detected respectively by clone formation assay and MTT cell proliferation assay.Results：Expression of TRIM28 in SK-MES-1 cells was inhibited significantly by RNA interference. The activity and growth of SK-MES-1 cells decreased obviously after the TRIM28 gene was inhibited.Conclusions：TRIM28 can promote the growth of lung squamous cell carcinoma SK-MES-1 cells.
