Cox-2在槲皮素抑制耐药白血病HL-60、HL-60A细胞增殖中的作用

摘　　要：目的探讨槲皮素体外对急性白血病耐药细胞株HL-60、HL-60A的抑制作用及环氧化酶-2(Cox-2)在其中起的作用。方法 CCK-8法检测槲皮素对HL-60及HL-60A细胞的抑制作用,流式细胞仪检查槲皮素诱导细胞凋亡作用;Westert-blot法检测Cox-2在正常人外周血单个核细胞、HL-60及HL-60A细胞表达情况;观察不同浓度槲皮素处理HL-60及HL-60A细胞前后Cox-2表达的变化趋势。结果槲皮素可以诱导HL-60及HL-60A细胞凋亡及抑制其增殖;Cox-2在正常人外周血单个核细胞低表达,在HL-60及HL-60A表达明显升高,HL-60A表达升高更加明显;经槲皮素处理后,Cox-2在HL-60及HL-60A的表达明显下降,HL-60A下降更加明显。结论Cox-2表达与急性白血病细胞耐药有关,槲皮素通过抑制Cox-2的表达诱导HL-60A细胞凋亡而抑制细胞增殖。Objective To investigate the inhibitory effect of Quercetin on leukemia HL-60 and HL-60 A cell lines in vitro and the the role of Cox-2plays during the reaction. Methods The CCK-8assay was used to measure the inhibition of Quercetin on HL-60 and HL-60 Acell lines,and a flow cytometry was used to detect the apoptosis of HL-60 and HL-60 Acells after exposure to Quercetin;the expressions of Cox-2in HL-60,HL-60 Acells and in peripheral blood mononuclear cells（PMBC）isolated from healthy adults were detected by using Western-blot.The change tendency of Cox-2expression in HL-60,HL-60 Acells were detected before and after treatment with Quercetin at different concentrations. Results Quercetin inhibited HL-60 and HL-60 Acell proliferation and induced apoptosis.Compared with healthy adult PBMC,low expression in PBMC,the expression of Cox-2on leukemia HL-60 and HL-60 Acells increased,and the expression of Cox-2on HL-60 Aincreased more significantly;the expression of Cox-2on HL-60 Adecreased after the cells were exposed to Quercetin,the HL-60 Acells decreased more significantly.Conclusion The expression of Cox-2was related to acute leukemia cells drug-resistance,Quercetin induced apoptosis by inhibiting the Cox-2expression for inhibiting HL-60 Acell proliferation.
