多奈哌齐对阿尔茨海默病患者调节性T细胞的影响

摘　　要：目的:观察阿尔茨海默病(AD)及认知功能正常的老年患者外周血CD4+CD25+CD127dim/-调节性T细胞比例及IL-10、TNF-α、TGF-β1及hs-CRP水平及多奈哌齐治疗1年后调节性T细胞比例的变化。方法:采用流式细胞术检测外周血CD4+CD25+CD127dim/-Treg细胞数量,酶联免疫法(ELISA)法测定血清IL-10、TNF-α、TGF-β1及hs-CRP水平。结果:1年后AD对照组及健康对照组CD4+CD25+CD127dim/-T细胞占CD4+CD25+T细胞比例均明显增加(P<0.05),多奈哌齐治疗组CD4+CD25+CD127dim/-T细胞占CD4+CD25+T细胞比例无明显变化。AD干预组、对照组和健康对照组间血清IL-10、TNF-α、TGF-β1及hs-CRP水平无明显变化,多奈哌齐治疗前后IL-10、TNF-α、TGF-β1及hs-CRP水平无明显变化。结论:外周血Treg水平在健康及AD老年人群均随增龄而增加,多奈哌齐的干预可以延缓AD患者Treg比例随增龄而增加。Objective：To investigate the frequency of CD4＋CD25＋CD127dim/-regulatory T cells in peripheral blood of the normal cognitive elderly and AD and the change of the Treg frequency with treatment of donepezil for one year.Method：The number of CD4＋CD25＋CD127dim/-Treg cell was detected by use of flow cytometry.The level of serum IL-10、TNF-α、TGF-β1and hs-CRP were determined by enzymelinked immunosorbent assay.Result：One year later the frequency of CD4＋CD25＋CD127dim/-T lymphocyte was high in the normal control group and AD control group（P〈0.05）.The serum levels of IL-10 、TNF-αand TGF-β1has no significant change.Conclusion：The frequency of Treg increases with age in both healthy elderly and AD patients.Donepezil can delay the increasing frequency of Treg in patients with AD.
