HSP70在先天性白内障小鼠晶状体上皮细胞中的表达及意义

摘　　要：目的探讨热休克蛋白(heat shock protein,HSP)70在先天性白内障小鼠模型晶状体上皮细胞中的表达及意义。方法先天性白内障BALB/c-Cat小鼠为实验组,正常BALB/c小鼠为对照组,每组16只。将实验组与对照组均各取8只分别进行Western blot蛋白印迹法和逆转录聚合酶链反应(RT-PCR)法检测小鼠眼睛晶状体上皮细胞中HSP70及HSP70基因的表达水平,以β-actin作为内参校正,比较2组间HSP70表达的差异。结果 HSP70在小鼠晶状体上皮细胞的相对表达值实验组为0.105 0±0.041 4,对照组为0.188 7±0.0236,2组比较差异有统计学意义(t=4.972 0,P<0.01);小鼠晶状体上皮细胞HSP70基因的相对表达值实验组为0.131 3±0.003 5,对照组为0.334 0±0.004 2,2组比较差异有统计学意义(t=37.026,P<0.01)。结论 HSP70在先天性白内障小鼠病情的发生、发展中能起一定的细胞保护作用。Objective It is to approach the expression of heat shock protein 70 （HSP70） in lens epithelial cells （LECs） in mice model of congenital cataract and clarify the effect of HSP70 on pathogenesis of congenital cataract. Methods The lens capsule membrane in mice was chose as the research object, there was 16 cases （32 eyes） of normal BALB/c mouse in control group, and 16 cases （32 eyes） of congenital cataract BALB/c -Cat mouse in experimental group. The expression of HSPT0 protein and HSPTO gene was assayed using Western blot method of protein imprinting and reverse transcription polymerase chain reaction （ RT - PCR） method in the two groups of mice eye lens epithelial cells. Using β - actin as the confidential ref- erence items for correction, the differences of HSP70 expression was compared between the two groups. Results The relative values of HSP70 expression to β - actin is O. 105 0 ＋ O. 041 4 in congenital cataract LECs, while is 0. 188 7 ＋ O. 023 6 in nor- mal control group, there was significant different in the expression of HSP70 between the two groups （ t = 4. 972 0, P 〈 0.01 ）. The relative expression value of HSP70 gene is 0. 131 3 ± 0. 003 5 in the experimental group and O. 334 0±0. 004 2 in the nor- mal group, there was significant difference between them （t = 37. 026, P 〈 0.01 ）. Conclusion HSP70 is associated with the occurrence and development of congenital cataract and it plays an important role in the cytoprotection.
