摘 要:目的探讨参附注射液对氯胺酮致老龄大鼠认知功能障碍的影响。方法健康雄性SD老龄大鼠40只,鼠龄16-24个月,体重350-450 g,随机分为C、S、K、SK组4组(n=10)。S组静脉输注参附注射液10 m L/kg,K组静脉输注氯胺酮40 mg/(kg·h),SK组静脉输注参附注射液10 m L/kg+氯胺酮40 mg/(kg·h),C组静脉输注等容量生理盐水,2 h/d,连续7 d。给药结束后测定大鼠认知功能,记录逃避潜伏期和穿越平台次数。认知功能测试完毕后处死大鼠,取海马组织,检测海马组织神经元凋亡情况,计算神经元凋亡率。用实时荧光定量PCR检测Fas基因及SABC法测定海马组织内Fas蛋白表达情况。结果与C组比较,S组逃避潜伏期、穿越平台次数、海马神经元凋亡率和海马组织内Fas mRNA相对表达量差异无统计学意义(P〉0.05),K组和SK组逃避潜伏期延长,穿越平台次数减少,海马神经元凋亡指数升高,Fas mRNA相对表达量上升(P〈0.05);与K组比较,SK组逃避潜伏期缩短,穿越平台次数增加,海马神经元凋亡指数降低,Fas mRNA相对表达量下降(P〈0.05)。结论参附注射液可改善氯胺酮所致老年大鼠认知功能障碍,其机制可能与参附注射液可下调Fas基因表达从而抑制氯胺酮诱发的细胞凋亡作用有关。Objective To investigate the effects of Shenfu injection on ketamine - induced cognitive dysfunction. Methods Forty SD male SD rats (aged 16 - 24 months, weighing 350 - 450 g) were randomly divided into four groups ( n = 10 per group) : Control group ( Group C), Shenfu group ( Group S), ketamine Group ( Group K) and Shenfu plus ketamine Group (Group SK). In Group S and K, Shenfu (10 mL/kg) and ketamine (40 mg/kg) were intravenously injected per day for 7 days, respectively ; while Shenfu plus ketamine were given to the subjects of Group SK and replaced saline were given to those in Group C. Morris water maze was used for evaluation of cognitive function after drug administration. Following the water maze test, the rats were sacrificed and the hippocampi were harvested for the determination of neuron apoptosis (TUNEL) and the expression of Fas mRNA and protein ( Real - time PCR and immuno - histochemistry). Results Compared to Group C, no significant difference was revealed in all results of the Morris water maze test of Group S ( P 〉 0. 05 ). However, in Group K and KS, significant prolonged escape latency, reduced crossing the platform count, increased neuron apoptosis and up - regulated Fas were observed in comparison with Group C ( P 〈 0. 05 ). Compared to Group K, significant shortened escape latency, increased crossing platform count, reduced neuron apoptosis and down - regulated Fas were observed in Group KS (P 〈 0. 05). Conclusion Shenfu injection attenuates the cognitive dysfunction induced by ketamine, with the possible mechanism related to the down - regulation the Fas gene expression as well as neuron apoptosis.
