摘　　要：高相色效液谱仪使用后,应使用适当的溶剂清洗色谱柱对使用缓冲盐类的流动相,用大比例的水彻底清洗其中的缓冲盐溶液,然后用甲醇或乙腈冲洗,并保持在甲醇或乙腈中。为了能够延长色谱柱的使用周期,对色谱柱的清洗时间适当的延长,即每日先用大比例的水(90∶10=水∶甲醇或乙腈)清洗约2 h,再用甲醇或乙腈清洗约2 h,结果色谱柱的使用周期明显延长,降低了实验室的检验成本。Column and flow system of High performance liquid chromatography need to be cleaned by an appropriate solvent after a test, especially for the column and flow system which has used moving phase of buffer salt solvent and stopped working for more than one night, must be cleaned by adequate water（large proportion of water, the specific proportion of water refer to the performance of different columns）and methanol or acetonitrile successively, the amount of water must be more than 20 times of bed volume, and then keep it filled with methanol or acetonitrile. There will be some strong reservations substance accumulate gradually in the column after a period of time that will not flow out with the mobile phase and affect the efficiency of the column, this is the main reason cause of the phenomenon such as baseline instability or ghost peaks on the chromatogram, what＇s more, reduce the column cycle. An effective way to solve this problem is to flush the column with a strong solvent. Aiming at this problem, in this paper, we researched The effect of liquid chromatography cleaning time on the C8、C18 reverse- phase chromatographic column using period.
