摘  要:目的通过探讨天津地区急性脑梗死发生与血管紧张素转化酶(ACE)rs4646994和rs35397082基因多态性及血浆浓度的相关性,找出天津地区脑梗死发生的危险因素,为预防脑梗死的发生提供相应的检测指标。方法用基因测序技术以及ELISA方法分别检测88例急性脑梗死患者和81例健康志愿者的ACE的单核苷酸多态性(SNP)以及血浆浓度。结果正常对照组中rs4646994的插入型SNP与急性脑梗病人组比较,差异无统计学意义。但是rs35397082的插入型SNP却只存在于脑梗死病人组中,比例高达40.91%,插入型的rs35397082与急性脑梗死的发生存在着显著的相关性(P<0.05)。同时病人组的ACE浓度明显高于正常对照组(P<0.001)。结论天津地区rs35397082的插入型似乎为脑梗死发生的一个相对独立的危险因素,同时血浆中ACE的浓度也与脑梗死的发生存在着一定的相关性,该初步研究结果可为今后预防天津地区的脑梗死发生提供一定的检验指标。Objective To study the relationship between the acute cerebral infarction and the angiotensin converting enzyme(ACE) gene(rs4646994 and rs35397082) polymorphisms and the plasma concentration of ACE in Tianjin area,so as to find the risk factors of cerebral infarction and provide corresponding detection indexes for prevention of cerebral infarction incidence.Methods Gene sequencing and ELISA method were used to detect the single nucleotide polymorphisms(SNPs)(rs4646994and rs35397082) and plasma level of ACE gene in 88 cases of acute cerebral infarction and 81 healthy volunteers.Results The difference of rs4646994 insertion type SNPs had no statistical significance between normal control group and acute cerebral infarction group.But the insertion type SNPs of rs35397082 was found only in acute cerebral infarction group,accounting for 40.91%,indicating the significant correlation between rs35397082 insertion type SNPs and acute cerebral infarction(P〈0.05).Plasma ACE level of the acute cerebral infarction patients(16.30 ±2.15) U/ml was much higher than the control subjects(9.89 ± 1.68) U/ml(P〈0.001).Conclusion The insertion type of rs35397082 seems a independent risk factor for occurrence of acute cerebral infarction in Tianjin area,and the plasma level of ACE also has close relationship with the acute cerebral infarction,which has provided some new risk factors of cerebral infarction in Tianjin area.
