益气养阴活血法对糖尿病模型大鼠PDGF-B和ICAM-1表达的干预作用研究

摘　　要：目的:探讨具有益气养阴活血作用的丹蛭降糖胶囊对模型大鼠血小板衍生生长因子-B(PDGF-B)和细胞间黏附分子-1(ICAM-1)的影响。方法:取大鼠55只,分为5组,除正常对照组外,其余各组先给予高脂饲料喂养4周,一次性腹腔注射链脲佐菌素(STZ)35 mg/kg,造模成功后的SD大鼠随机分为4组:丹蛭降糖胶囊高剂量组(DJC高)、丹蛭降糖胶囊低剂量组(DTC低)、吡格列酮组、模型组。除模型组和正常对照组外,药物组连续灌胃8周,取血测定血糖和PDGF-B(血小板衍生因子)和ICAM-1(细胞黏附分子)(酶联免疫吸附法)。结果:模型组血清中PDGF-B、ICAM-1明显升高,与正常组比较有显著统计学意义(P<0.01);予丹蛭降糖胶囊治疗后,两指标明显下降(P<0.05,P<0.01),优于吡格列酮组(P<0.05)。单用丹蛭降糖胶囊血糖明显下降,与模型组比较差异显著(P<0.01);丹蛭降糖胶囊组与吡格列酮组比较,无明显差异(P>0.05)。结论:丹蛭降糖胶囊可显著降低糖尿病大鼠PDGF-B和ICAM-1的高表达,降低血糖,改善模型大鼠精神萎靡、毛发稀疏无光、活动减少的状态。Objective： To investigate the influence of Danzhi jiangtang capsule（ 丹蛭降糖胶囊） which has the function of supplementing qi,nourishing yin and activating blood circulation on diabetic model rats＇ platelet- derived growth factor- B（ PDGF- B） and intercellular adhesion molecule- 1（ ICAM- 1）. Methods： 55 rats were divided into 5 groups,feed the rats with high- fat diet except the normal group,after 4 weeks,give diabetic model rats intraperitoneal injection with streptozotocin（ STZ） 35 mg / kg once only,randomly divided successful molding rats into high dose Danzhi jiangtang capsule（ DJC） group,low dose Danzhi jiangtang capsule（ DJC） group,Pioglitazone group and model group. Normal group was regard as control group. Rats were given intragastric administration continuously for 8 weeks except model group and normal group,drawn their blood and measured their blood glucose level,blood fat level,PDGF- B and ICAM- 1. Results： Serum PDGF- B in the model group increased significantly,compared with the normal group（ P〈0. 01）; after given Danzhi jiangtang capsule,the index decreased significantly（ P〈0. 01）,that was better than pioglitazone group. The ICAM- 1index was significantly increased in model group rats（ P〈0. 01）,suggesting that the model was successful.ICAM- 1 index of high DJC group decreased significantly after 8 weeks＇ intragastric administration of drugs,compared with model group（ P〈0. 01）. Conclusion： Danzhi jiangtang capsule could significantly decrease the high expressions of PDGF- B and ICAM- 1,decrease blood glucose,improve the model rats＇ state such as depression,thinning hair and activity reduced.
