腺病毒E1对人黑色素瘤靶向性治疗探讨

摘　　要：目的 探讨腺病毒E1对人黑色素瘤靶向性治疗效果.方法 PCR克隆获取腺病毒E1区,增强子DNA片段及鼠酪氨酸酶启动子,采用基因重组法进行构建靶向人黑色素瘤的选择复制性腺病毒的穿梭载体;同源重组法获取人黑色素瘤的选择复制性腺病毒E1,腺病毒E1小剂量攻击人黑色素瘤细胞系SK-Mel-I,同时攻击人肝细胞癌细胞系HepG2,对细胞形态发生的改变、细胞存活数进行记录,利用RT-PCR检测E1A表达,证实腺病毒E1特异性复制.结果 PCR检测结果显示,SK-Mel-I对腺病毒E1裂解杀伤敏感,但是对于HepG2不敏感,提示腺病毒E1在人黑色素瘤细胞中可特异地复制.结论 腺病毒E1对人黑色素瘤具有很好的靶向性.Objective To investigate the adenovirus E1 mediated targeting treatment on human melanoma.Methods Adenovirus E1 region,enhancer DNA fragment and the murine tyrosinase promoter were obtained by PCR amplication.Replication adenovirus shuttle vector was constructed by gene recombination.Human melanoma cell line SK-Mel-I and human Liver cancer cell line hepG2 were respectively transfected by adenovirus E1.Cell morphology and cell viable count were recorded and E1A expression was validated by RT-PCR method.Results SK-Mel-I was sensitive to the cleavage of adenovirus E1 and HepG2 was not sensitive,which suggested that the adenovirus E1 could replicate in human melanoma cell.Conclusions Adenovirus E1 has good targeting treatment on the human melanoma.
