摘 要:目的口腔鳞状细胞癌是头颈部常见的恶性肿瘤,约占全身恶性肿瘤的3%。口腔鳞癌的淋巴转移是影响患者预后的重要因素。淋巴系统如何促进了肿瘤的播散,其具体迁移及侵袭机制一直不明确。基质金属蛋白酶系统在肿瘤从原位的增殖性肿瘤到具有侵袭、转移特性的癌的演变过程中起着重要作用。本实验初步研究了不同转移特性的癌细胞系及口腔鳞癌的组织病理切片中MMP-1、MMP-2、MMP-3和MMP-7的表达。方法通过Transwell、RT-PCR、Westernblot、免疫细胞化学法、免疫组织化学法研究不同转移特性的细胞系GDC185、Tca8113、Tca83及口腔鳞癌组织病理切片中基质金属蛋白酶(MMP-1、MMP-2、MMP-3、MMP-7)在m RNA和蛋白质水平的表达。结果 1Westernblot显示在三种不同特性的口腔鳞癌细胞系中MMP-1、MMP-2、MMP-3在Tca83中表达较GDC185、Tca8113高,但是其差异无统计学意义;2Westernblot显示在三种不同特性的口腔鳞癌细胞系中MMP-7在Tca83中表达较GDC185、Tca8113高,其差异具有统计学意义;3免疫组化结果显示MMP-7在高分化口腔鳞癌组织中表达较高,其差异具有有统计学意义。结论基质金属蛋白酶MMP-7在口腔鳞状细胞癌中的表达和癌的分化级别有一定的相关性。Objective Oral squamous cell carcinoma (OSCC) is a common kind of malignant tumor at the head and neck region and its incidence accounts for 3% of that of all the malignant tumors of the body. The lymphatic metastasis of OSCC is an important factor that can influence the prognosis of a patient. How the lymphatic system expedites the spread of this kind of tumor and the specific migration and aggression mechanisms are still not clear. The matrix metalloproteinase system plays an important role in the process during which the tumor evolves from a kind of proliferative tumor in situ to a kind of aggressive and metastatic carcinoma. This study preliminarily investigated the expression of MMP-1, MMP-2, MMP-3 and MMP-7 in the histopathologic sections and cell lines of OSCC with different metastatic features. Methods The expression at the mRNA and protein level of matrix metalloproteinase (MMP-1, MMP-2, MMP-3 and MMP-7) in the histopathologie sections, cell lines GDC185, Tca8113, Tea83 were investigated through the Transwell, RT-PCR, Westemblot, and immunocytochemical and immunohistochemical approaches. Results (1)There were no statistically significant differences in the expression of MMP-1, MMP-2 and MMP-3 in three kinds of OSCC cell lines with different features by westernblot. (2)By westmblot , the expression of MMP-7 was higher in the OSCC cell line Tea83, which is more aggressive and with less tumorigenesis and lymph node metastasis. Its expression was lower in OSCC cell lines GDC185 and Tca8113, which are less aggressive and with more tumorigenesis and lymph node metastasis. However, the differences are statistically significant. (3)The expression of MMP-7 was higher in highly differentiated OSCC tissues and the differences were statistically significant in immunohistochemical. Conclusion The expression of MMP-7 is correlated to the differentiation level of OSCC.
