摘  要:目的研究青霉素腹腔注射制作大鼠癫痫模型的方法。方法 36只Wistar雄性健康大鼠,随机分为正常对照组、癫痫模型脑电图组、癫痫模型单位放电组,每组各12只。癫痫模型组用青霉素钠按6×106 U/kg腹腔注射,观察癫痫发作级别,观察脑电图变化,记录海马神经元单位放电。结果正常对照组大脑皮质脑电以α波为主要表现,频率为7~9 Hz,无明显阵发性节律出现,波幅〈50μV。癫痫模型脑电图组癫痫波包括尖波、慢波、尖慢波、棘波、棘慢波等。正常对照组大鼠共记录到24个单位海马神经元放电,放电频率以中低频放电为主,放电形式以单个不均匀放电为主;癫痫模型单位放电组共记录到78个单位海马神经元放电,放电频率以高频放电为主,放电形式则以混合型放电为主;与正常对照组比较,海马神经元单位放电数、放电频率及放电形式差异均有统计学意义(P〈0.05)。结论青霉素癫痫模型制作成功后,在额叶皮质记录到各种癫痫放电,海马神经元单位放电数明显增多,放电频率高,并以混合型暴发放电为主。Objective To investigate the method of constructing epilepsy model in rats by intraperitoneal injection of Penicillin. Methods Thirty-six male and healthy Wistar rats were divided into 3 groups: normal control group(n = 12), electroencephalogram (EEG) of epilepsy model group(n = 12) and unit discharge of epilepsy model group (n = 12). The epilepsy model group received intraperitoneal injection of Penicillin sodium at 6 x 106 U/kg, the seizure grade and change of EEG were observed, and hippocampal neuronal unit discharge was recorded. Results The main performance of cortical EEG in normal control group was α-wave with frequency of 7 - 9 Hz, no obvious paroxysmal rhythmic was found and the amplitude was lower than 50 μV. The epilepsy ECG waves in epilepsy model group included sharp-wave, slow-wave, sharp-slow-wave, spike-wave and spike-slow-wave. Twenty four units of hippocampal neurons discharge were recorded in normal control group, which were mainly in low-middle frequency and uneven single discharge. Seventy-eight units of hippocampal neuron discharge were recorded in epilepsy model group, which were mainly in high frequency and mixed discharge. The differences in unit discharges of hippocampal neuron, frequency and discharge form were significantly different between epilepsy model group and normal control group(P 〈 0.05). Conclusion It is demonstrated that the Penicillin epilepsy model is successfully constructed, various patterns of epilepsy discharge is recorded in the frontal cortex, and the hippocampal neuron discharge number is increased with high frequency, mainly in the form of mixed- burst discharge.
