摘  要:目的:构建核素标记自主合成九环肽DTPA-c(CGRRAGGSC)的生物分子靶向探针,研究其在肝癌体内分布及细胞定位。方法:Western blot检测人肝癌细胞MHCC97H及人正常肝细胞7702白介素11受体(IL-11R)表达情况;进行标记探针与MHCC97H细胞荧光定位;确定99Tcm标记DTPA-c(CGRRAGGSC)最佳条件;观察标记探针99Tcm-DTPA-c(CGRRAGGSC)在MHCC97H肝癌模型鼠体内生物分布。结果:IL-11R表达量MHCC97H细胞是7702细胞的21倍;荧光实验证实DTPA-c(CGRRAGGSC)与MHCC97H细胞膜和胞浆特异性结合;99Tcm-DTPA-c(CGRRAGGSC)标记率>98%;瘤体给药后进行ECT显像,证实具有良好靶向性及稳定性。结论:核素及荧光标记九环肽可靶向特异性结合肝癌MHCC97H肿瘤模型。Objective: To built a nuclide biological target probe and assess the biostribution in subcutaneous transplanted model of MHCC97H in nude mice. Methods:The expression changes of IL-11R in MHCC97H tumors and 7702 nomal hepatic cell were examined by Western blot. The binding site of the molecular target and internalization with MHCC97H cells was observed under a fluorescence microscope. DTPA-c( CGRRAGGSC) was radiolabeled with 99Tcm. The biodisrtibution in nude mice bearing MHCC97H tumors after injection of 99Tcm-DTPA-c( CGRRAGGSC) molecular probe was obsored. Results: Western blot showed the expression of IL-11R in MHCC97H cells was 21 times than that of 7702 cells. The c ( CGRRAGGSC) possessed the ability of specific binding with cytomembrane of MHCC97H cells. The labeling rate of 99Tcm-DTPA-c(CGRRAGGSC) was more than 98%. ECT imags confirmed good targeting and stability of 99Tcm-DTPA-c(CGRRAGGSC) probe in nude mice bearing MHCC97H. Conclusion:Nuclide marked DTPA-c(CGRRAGGSC) has specific binding to the MHCC97H tumor model of liver cancer.
