肾透明细胞癌QKI、HIF-1α的表达及相关性研究

摘  要:目的研究QKI、HIF-1α在人肾透明细胞癌中的表达及其相关性,探讨QKI、HIF-1α的表达与肾透明细胞癌病理分级、浸润深度及临床分期的关系。方法收集110例肾透明细胞癌和10例正常肾组织石蜡包埋标本作为研究对象,采用免疫组化SABC法检测QKI、HIF-1α的表达和分布,并应用图象分析仪判定,统计学处理,对肾透明细胞癌的病理分级、浸润深度及临床分期进行对比分析及相关性研究。结果 QKI在肾透明细胞癌中的阳性表达率为67.3%,正常肾组织为100%,差异有统计学意义(P<0.05);HIF-1α在肾透明细胞癌中的阳性表达率为92.7%,正常肾组织为30%,统计学有显著性差异(P<0.01)。在肾透明细胞癌中QKI的表达强度与病理分级(r=-0.471,P<0.001)、浸润深度(r=-0.392,P<0.001)、TNM分期(r=-0.451,P<0.001)均为线性负相关;HIF-1α的表达强度与病理分级(r=0.404,P<0.001)、浸润深度(r=0.324,P=0.001)、TNM分期(r=0.330,P<0.001)均为线性正相关。QKI与HIF-1α表达强度具有统计学差异(P=0.002),并呈线性负相关性(r=-0.417,P=0.002)。结论 QKI的表达与肾透明细胞癌病理分级、浸润深度及临床分期呈负相关性,而HIF-1α的表达与其呈正相关性,在发病机制中可能涉及到QKI、HIF-1α的异常表达。同时,作为新的分子标志物,QKI、HIF-1α可作为判定肾透明细胞癌恶性程度、进程及预后的一项有价值的生物学指标,对临床诊治也可提供有意义的帮助。This study performed to explore the associativity between QKI and HIF-1α expressions in human renal clear cell carcinoma,for approaching their relations to the pathological grading,invasion severity and clinical staging of renal clear cell carcinoma. Total of 110 human renal clear cell carcinoma tissues and 10 normal human renal tissues were prepared by paraffin embedding. Levels of QKI and HIF-1α in the samples were detected by immunohistochemistry SABC method,and then the samples were analyzed by using image pattern analyzer and statistic methods for the comparative analysis and correlation research of pathological grading,invasion severity and clinical staging of renal clear cell carcinoma. The immunohistochemistry results showed the expression of QKI in the renal clear cell carcinoma tissues and the renal normal tissues were 67.3% and 100% respectively(P 0.05),while the expression of HIF-1α in the renal clear cell carcinoma tissues and normal renal tissues were 92.7% and30% respectively(P 0.01). And the statistic analysis found that the expression of QKI in the renal cell carcinoma showed a significant negative linear correlation with pathologic grading( r =-0.471,P 0.001),depth of invasion(r =-0.392,P 0.001) and TNM classification(r =-0.451,P 0.001). While the expression of HIF-1α in renal clear cell carcinoma showed a positive linear correlation with pathologic grading(r=0.404,P 0.001),depth of invasion(r= 0.324,P=0.001) and TNM classification(r=0.330,P 0.001). The expression of QKI and HIF-1α in renal clear cell carcinoma had significant difference(P =0.002),and there was significant negative linear correlation between them(r=-0.417,P=0.002). All the results come to the conclusion that QKI and HIF-1α are involved in theoccurrence and development of renal clear cell carcinoma. The abnormal expression of QKI and HIF-1α may participate in the pathological mechanism ofrenal clear cell carcinoma. Meanwhile,as novel biomarkers,QKI and HIF-1α can be used to est imat
