摘 要:目的观察转化生长因子β1(TGF-β1)和Smad7在乳腺癌组织内的表达,分析与乳腺癌进展及淋巴结转移之间的关系。方法取乳腺癌病例52例,其中,淋巴结转移组32例,无淋巴结转移组20例。应用免疫组化法和Western blot技术观察TGF-β1和Smad7在乳腺癌组织内的表达。结果 TGF-β1主要表达于乳腺癌细胞胞浆内,与乳腺癌临床分期和淋巴结转移之间具有相关性,其在淋巴结转移组的表达率明显高于无淋巴结转移组(P=0.021)。Smad7表达于乳腺癌细胞胞浆和胞核内,与乳腺癌临床分期及淋巴结转移密切相关,在淋巴结转移组的表达率明显高于无淋巴结转移组(P=0.023)。TGF-β1与Smad7在乳腺癌组织内的表达呈显著的正相关(P=0.016)。结论 TGF-β1和Smad7的表达与乳腺癌的分期和淋巴结转移密切相关,表明TGF-β1/Smad7信号通路可能在乳腺癌进展和淋巴结转移过程中起重要作用。Objective To investigate the effects of transforming growth factor β1 (TGF- β1) and Smad7 in lymph node metastasis. Methods Fifty-two paraffin-embedded specimens from patients with breast cancer were divided into lymph node metastatic group(n=32) and nonmetastatic group(n=20). The expressions of TGF-β1 and Smad7 were detected by immunohistochemical stain and Western blot. Results TGF-β1 product was localized in the cytoplasm of the tumor cells, the expression level of TGF- β1 was closely correlated with clinical stage and lymph node metastasis, higher in lymph node metastatic group than in nonmetastatic group(P=0.021). Smad7 product was observed in cytoplasm and nucleus of tumor cells, the overexpression of Smad7 was related to clinical stage and lymph node metastasis in breast cancer, higher in lymph node metastatic group than in nonmetastatic group(P =0.023). TGF-β1 expression was positive correlated with Smad7 expression(P=0.016). Conclusion TGF- β1 and Smad7 expression was positively correlated with clinical stage and lymph node metastasis in breast cancer, which indicates that TGF- β1/Smad7 signal pathway may play an important role in tumor progress and lymph node metastasis in breast cancer.
