摘  要:目的:研究幽门螺杆菌(Hp)感染对胃黏膜G、D细胞及胃泌素(GAS)、生长抑素(SS)表达的影响。方法:Hp标准菌SS2000菌液灌胃C57BL/6小鼠构建Hp感染小鼠慢性胃炎动物模型,免疫组化染色观察小鼠胃黏膜G、D细胞的改变,图像分析软件(Image-Pro-Plus,IPP)分析GAS、SS阳性灰度值,Real time-PCR检测小鼠胃窦黏膜GAS mRNA和SS mRNA相对表达量。结果:与空白组比较,模型组G细胞数目、GAS灰度值、GAS m RNA表达水平显著增高(P<0.05),D细胞数目、SS灰度值、SS mRNA表达水平无明显改变。结论:Hp感染可引起胃黏膜GAS mRNA表达上调,胃窦黏膜G细胞增多,促进GAS分泌,可能是胃黏膜病理改变加重的原因。Objective: To investigate the influence of Helicobacter pylori (lip) infection on gastric mucosal G cells and D cells, and the expression of gastrin (GAS) and somatostatin (SS) mRNA in mice. Methods: SPF C57BL/6 mice were given Hp SS2000 by gavage to construct animal model of chronic gastritis. Adopting immunohistochemistry starting and image-pro-plus (IPP) to detect the ex- pression and localization of GAS and SS in gastric antrum tissue, and analyzing the changes on G cell and D cell. The GAS and SS mRNA expression levels in gastric mucosa were detected before and after Helicobacter pylori infected by real time PCR. Results: Comparing with the blank group, model group's number of G cells, GAS gray value, GAS expression level of mRNA was significantly increased (P 〈 0.05) ; the number of D cells, SS gray value, SS mRNA expression level of model group did not sig- nificant changed ( P 〉 0.05 ). Conclusions: The infection of Hp can upregulate the expression of GAS mRNA in gastric mucosa, and increase the amount of G cells in gastric mucosa, and promote the se- cretion of GAS. It may be a cause of gastric mucosal pathological change aggravation.
