转录因子E2F1在脑膜瘤中的表达和临床意义

摘　　要：目的研究转录因子E2F1在脑膜瘤中的表达情况及其临床意义。方法选择57例经病理确诊为脑膜瘤患者及同期行脑外科手术治疗的38例非脑膜瘤患者,应用实时定量PCR方法和Western blot方法定量检测转录因子E2F1在脑膜瘤组织和正常脑膜中的表达情况,并采用免疫组织化学染色方法检测脑膜瘤组织中E2F1蛋白的表达定位。根据E2F1的表达情况分析其与脑膜瘤分型和预后的关系。结果实时定量PCR结果显示脑膜瘤组织中E2F1 mRNA表达水平显著高于正常脑膜组织,而且恶性脑膜瘤组织中E2F1 mRNA表达高于良性脑膜组织(P<0.01),Western blot结果也证实脑膜瘤组织中E2F1蛋白表达水平显著高于正常脑膜组织(P<0.01)。免疫组织化学染色结果显示脑膜瘤组织中E2F1蛋白主要定位在细胞核中。脑膜瘤组织中E2F1强阳性表达率为65%,正常脑膜组织中E2F1强阳性表达率为10%,二者比较差异具有统计学意义(P<0.01)。不典型和恶性脑膜瘤组织中E2F1强阳性表达率显著高于良性脑膜瘤。3年内脑膜瘤复发组E2F1表达阳性率显著高于无复发组(P<0.01)。结论 E2F1在脑膜瘤组织中表达增强,并且与脑膜瘤的恶性程度和复发率呈正相关。Objective It is to observe the expression of transcriptional factor E2F1 and its clinical pathological significance in meningiomas. Methods 57 cases of meningiomas tissues diagnosed by pathological examination were included in this study,38 cases of normal meninges tissues were used for control. Real-time PCR and Western blot were adopted for the quantification of E2F1 expression in meningiomas,immunohistochemistry was used to detect the E2F1 protein localization in meningiomas.The potential role of E2F1 in tumor classification and prognosis were analyzed. Results Real-time PCR results showed that E2F1 mRNA level was significantly higher than normal meninges tissues. Western blot results showed that E2F1 protein level was significantly higher than normal meninges tissues（ P〈0. 01）. Immunohistochemistry results revealed that E2F1 protein was mainly localized at nuclei meningiomas（ P〈0. 01）. The positive expression rate of E2F1 in meningiomas was 65 %,which was significantly higher than normal meninges tissues（ P〈0. 01）. In addition,the positive expression rate of E2F1 in atypical and malignant meningiomas was 73%,which was significantly higher than benign meningiomas（ P〈0. 01）. The positive expression rate of E2F1 in meningiomas recurrence group were markedly higher than non recurrence group（ P〈0. 01）.Conclusion E2F1 is highly expressed in meningiomas,and is positively correlated with malignancy grade and recurrence of meningiomas.
