摘 要:[目的]研究苹果多酚对糖尿病小鼠血脂、血糖水平和肝、肾、脂肪组织PPARγmRNA表达的影响。[方法]雄性小鼠分为对照组(n=12)、模型组(n=12)和苹果多酚干预组(n=36)。通过给予高脂高糖饲料喂养,腹腔注射链脲佐菌素,建立糖尿病小鼠模型。应用苹果多酚对糖尿病小鼠干预治疗60 d。在试验结束时,集中处死鼠,搜集血标本和肝、肾、脂肪组织标本。检测各组大鼠的血MDA、FBG、FINS、TG、TC等浓度。采取RT-PCR方法,检测各组小鼠肝、肾、脂肪组织PPARγmRNA的表达。[结果]与Con组相比,模型组小鼠血清MDA、血脂、血糖、胰岛素水平显著升高(P<0.01),与模型组相比,苹果多酚能显著降低小鼠的血清MDA、血脂、血糖、胰岛素水平(P<0.05)。与Con组相比,模型组小鼠肝、肾、脂肪组织PPARγmRNA表达水平显著下降,苹果多酚组小鼠肝、肾、脂肪组织PPARγmRNA的表达水平较模型组均有明显增加(P<0.05)。[结论]苹果多酚能够降低糖尿病小鼠血清MDA、血脂、血糖水平,减轻胰岛素抵抗,提高PPARγmRNA的表达。苹果多酚可能发挥着类似PPARγ激动剂的效用,通过提高PPARγmRNA的表达来调节糖尿病小鼠的血糖、血脂代谢。[Objective] To study effects of apple polyphenol on blood lipid,blood glucose level of diabetic mice,liver,kidney,fat tissue PPARγ mRNA expression. [Method] Male mice were divided into Con group( n = 12),model group( n = 12),apple polyphenol group( n =36). Through feeding with high fat and high glucose diet,intraperitoneal injection of streptozotocin,diabetic mice model was established. Apple polyphenol was used to cure diabetic mice for 60 days. At the end of experiment,mice were killed to collect blood,liver,kidney,fat tissue samples. MDA,FBG,FINS,TG,TC concentration of mice in each group were detected. RT- PCR method was used to detect PPARγmRNA expression of liver,kidney,fat tissue. [Result] Compared with Con group,serum MDA,blood liquid,glucose,insulin level of mice in model group obviously increased( P 0. 01),compared with model group,apple polyphenol can significantly reduce serum MDA,blood liquid,glucose,insulin level( P 0. 05,P 0. 01). Compared with Con group,liver,kidney,fat tissue PPARγ mRNA expression level of mice in model group significantly decreased. [Conclusion] Apple polyphenol can reduce serum MDA,blood lipid,blood glucose level,insulin resistance,improve PPARγ mRNA expression. Apple polyphenol may have similar function of PPARγ agonist,which can adjust blood glucose,lipid metabolism through improving PPARγ mRNA expression.
