摘  要:以十字花科黑腐病菌(Xanthomonas campestris pv.campestris,Xcc)8004菌株为研究材料,采用双向电泳-质谱技术鉴定Hrc V控制的分泌蛋白质组。结果表明,野生型和突变体菌株均获得分辨率高且具有可重复性的双向电泳银染图谱,经过图像软件分析找出差异点61个,以野生型菌株为对照,22个蛋白点上调,39个蛋白点下调。挖取下调蛋白质点39个做质谱分析,质谱成功鉴定出25蛋白质,主要包括各种酶类、转运蛋白、延伸因子、Dna K蛋白、分子伴侣及假定蛋白。推测这些蛋白质可能在Xcc 8004的致病性中起到重要作用,为进一步研究Xcc的致病机理奠定基础。In the present study,two-dimensional electrophoresis( 2-DE) and mass were employed to analyze the secreted proteins controlled by Hrc V of Xanthomonas campestris pv. campestris( Xcc 8004). The results showed that the high quality and repetitive of 2-DE maps were obtained. Analysis through image analysis system indicated that there were 61 differentially expressed proteins between wild-type 8004 and hrc V mutants,including 22 up-regulated and 39 down-regulated proteins. The 39 down-regulated spots were collected and analyzed by MALDI-TOF-MS and Mascot software. As a result,25 spots were successfully identified,mainly including energy metabolism proteins,transporters,elongation factor,Dna K protein,chaperonin and hypothetical proteins. It is speculated that these genes might play important roles in virulence of Xcc 8004. The further analysis of these secreted proteins should make benefit for study on Xcc pathogenesis.
