摘 要:目的探讨NDRG2(N-myc downstream regulated gene 2)基因启动子甲基化与胃癌发生、发展的关系。方法取40例手术的胃癌组织及40例相应癌旁正常组织(距癌5 cm以上),MSP法检测其启动子区甲基化状况,免疫组化检测蛋白的表达水平。结果胃癌组织中NDRG2甲基化率为72.5%(29/40),而癌旁正常组织中仅8例(20.0%)发生了甲基化,差异有统学意义(χ2=22.17,P<0.01)。NDRG2甲基化与性别、年龄、TNM分期及有无淋巴结转移无相关性(P>0.05)。NDRG2蛋白在癌旁正常组织中的表达阳性率为87.5%(35/40),胃癌组织中为10.0%(4/40),差异有统计学意义(χ2=48.08,P<0.01)。结论 NDRG2基因可能参与胃癌的发生、发展。Objective To investigate the relationship between methylation status of the NDRG2(N-myc downstream regulated gene 2) gene and gastric cancer. Methods Gastric cancer tissues and normal cancer adjacent tissues were obtained surgically from 40 primary gastric cancer patients. Methylation-specific PCR was used to detect the methylation status of the NDRG2 genes and immunohistochemistry was used to detect the expression of NDRG2 protein in the above samples. Results Methylation rate of NDRG2 was 72. 5%(29 /40) in gastric cancer tissues and 20. 0%(8 /40) in normal cancer adjacent tissues,there was significant difference between two groups( χ^2= 22. 17,P 〈0. 001). Methylation rate of NDRG2 was not related with age,sex,TNM stage and lymph node metastasis. Expression of NDRG2 protein in gastric tissue was significantly lower than that in normal cancer adjacent tissues( χ^2= 48. 08,P 〈0. 01). Conclusion The promoter methylation of the NDRG2 genes may be associated with the occurrence of gastric cancer.
