PDX1在胃癌中的表达及与临床病理之间的关系

摘　　要：目的:探讨胰十二指肠同源基因1(pancreatic duodenal homeobox gene 1,PDX1)表达水平与胃癌临床病理之间的关系。方法:应用RT-PCR法分别测定AGS、BCG823、SGC7901等三株胃癌细胞中PDX1mRNA的表达,免疫印迹法测定胃癌组织芯片中PDX1蛋白表达水平。构建PDX1表达载体,转染胃癌细胞,转染后应用免疫印迹法测定胃癌细胞中PDX1、胃癌增殖相关蛋白(PCNA、Ki-67)和凋亡蛋白(Caspase3)的表达情况,并对胃癌细胞增殖及凋亡情况进行分析。结果:胃癌组织芯片中包括腺癌110例,黏液腺癌18例,印戒细胞癌15例,正常胃黏膜组织10例。正常胃黏膜组织中PDX1蛋白表达呈高水平,而在胃癌组织中PDX1蛋白表达水平下降或缺失(P<0.05)。PDX1表达水平与淋巴结转移及肿瘤分化程度有关(P<0.05)。PDX1表达载体经转染后,PDX1表达水平升高(P<0.05),而PCNA、Ki-67表达水平下降(P<0.05),Caspase3表达水平上调(P<0.05)。结论:PDX1在胃癌组织中表达下调与胃癌分化程度及淋巴结转移密切相关,其表达水平下调可能在胃癌病情进展中起到重要的作用。Objective：To investigate the expression levels of pancreatic duodenal homeobox gene 1（pancreatic du-odenal homeobox gene 1,PDX1）of proliferation and apoptosis on gastric cancer cell. Methods：The expression of PDX1mRNA of AGS,BCG823,SGC7901 were determented with Western blotting and RT - PCR. The PDX1 protein expression of gastric cancer tissue microarray were determented with immunohistochemistry. PDX1 expression vector construct were transfected into gastric cancer cells transfected with RT - PCR method,gastric proliferation - related proteins（PCNA,Ki - 67）and apoptotic proteins（Caspase3）expression,and gastric cancer cell proliferation and ap-optosis were analyzed. Results：Gastric adenocarcinoma tissue microarray 110 cases,mucinous adenocarcinoma 18 ca-ses,15 cases of signet ring cell carcinoma,10 cases of malignant stromal tumor of PDX1 protein expression was high, and in gastric cancer decreased or absent（P 〈 0. 05）. PDX1 expression was related lymph node metastasis and tumor differentiation（P 〈 0. 05）. After PDX1 expression vector transfected,PDX1 expression levels increased（P 〈 0. 05）, and PCNA,Ki - 67 expression levels decreased（ P 〈 0. 05）,whereas the expression level Caspase3 raised（ P 〈0. 05）. Conclusion：PDX1 downregulated is closely related with differentiation and lymph node metastasis of gastric cancer in gastric cancer,its lower expression level may play an important role in the progression of gastric cancer.
