摘 要:目的探讨新型抗癫痫药物唑尼沙胺的作用机制。方法将50只戊四氮致痫大鼠随机抽取8只为正常对照组,余42只造模成功后,随机抽取24只作为实验动物,随机分为唑尼沙胺组、苯巴比妥组和癫痫模型组,每组8只。监测4组大鼠唑脑组织NO、MDA含量及NOS、SOD的活性变化并观察大鼠海马组织结构的改变。结果癫痫组大鼠脑组织NO、MDA含量及NOS活性与正常对照组比较明显增高(P<0.05),SOD活性明显降低(P<0.05);唑尼沙胺组及苯巴比妥组大鼠脑组织的NO、MDA含量及NOS活性与癫痫组比较明显降低(P<0.05),SOD活性明显升高(P<0.05);唑尼沙胺能够改善癫痫大鼠海马组织的结构。结论唑尼沙胺通过降低脑组织NO含量而发挥其抗癫痫作用。Objective To investigate the action mechanism of zonisamide as a new antiepileptic drug. Methods Twenty-four rats with epilepsy induced by pentrazol were randomly divided into three groups: zonisamide group,phenobarbital group,epilepsy model group,with 8 rats in each group,at same time,the other 8 normal rats were used as control group. The contents of NO,MDA and activities of NOS,SOD in brain tissues of the rats were detected for the four groups,meanwhile,the changes of hippocampus tissue structure of the rats were observed. Results The contents of NO,MDA and activity of NOS were significantly increased in epilepsy model group,as compared with those in control group( P〈0. 05),however,the activity of SOD was significantly decreased in epilepsy model group( P〈0. 05). The contents of NO,MDA and activity of NOS were significantly decreased in zonisamide group and phenobarbital group,as compared with those in epilepsy model group,however,the activity of SOD was significantly increased in both groups( P〈0. 05). Furthermore,zonisamide could improve the structure of hippocampus tissue in rats with epilepsy. Conclusion Zonisamide plays an antiepileptic role by reducing the contents of NO in rats with epilepsy.
