摘  要:目的检测RECK基因(reversion-inducing-cysteine-rich protein with Kazal motifs)及基质金属蛋白酶-2(MMP-2)在不同卵巢癌细胞株SKOV-3、A2780、HO-8910及正常卵巢上皮细胞中的表达,并探讨其表达差异及其意义。方法应用RT-PCR检测卵巢癌细胞株SKOV-3、A2780、HO-8910及正常卵巢上皮细胞中RECK及MMP-2 m RNA的表达;应用Western blot方法检测不同卵巢细胞株中RECK蛋白的表达。结果卵巢癌细胞株SKOV-3、A2780、HO-8910中RECK m RNA的表达较正常卵巢上皮细胞中明显降低(P<0.01),MMP-2 m RNA表达则明显升高(P<0.01),但三种卵巢癌细胞株中RECK m RNA的表达没有差异(P>0.05);SKOV-3、A2780、HO-8910细胞株中RECK蛋白的表达量较正常卵巢上皮细胞明显降低(P<0.01)。结论 RECK基因在卵巢癌中的表达量明显下降,而MMP-2的表达量明显升高,RECK基因可能通过抑制MMP-2的表达起到抑癌基因的作用。Objective:To investigate the expression and significance of RECK gene and MMP-2 in the normal ovarian cell and ovarian cancer cell strains such as SKOV-3,A2780 and HO-8910.Methods:The m RNA level of RECK and MMP-2 in the ovarian cell strains was detected by techniques of reverse transcription polymerase chain reaction. The protein level was evaluated by Western blot. Results:The m RNA level of the RECK gene in the ovarian cell strains(SKOV-3,A2780 and HO-8910)was lower than that in the normal ovarian cell,but The m RNA level of the MMP-2 gene was higher. There is no difference in the m RNA level of the RECK gene among three ovarian cell strains. The protein of RECK in SKOV-3,A2780 and HO-8910 was lower than that in the normal ovarian cell. Conclusion:Our findings support the hypothesis that the RECK is a kind of tumor suppressor gene,which may act by inhibiting MMP-2 activation.
