摘  要:目的探讨牛磺酸锌(taurine-zinc,TZC)对铅暴露大鼠脂质过氧化损伤的拮抗作用。方法将40只3周龄健康雄性SD大鼠随机分成5组,分别为对照组、染铅组和10、50、100 mg/kg TZC拮抗组,每组8只。除对照组外,其余4组大鼠灌胃乙酸铅溶液(30 mg/kg)染毒;之前1 h,各剂量TZC拮抗组灌胃相应剂量的TZC溶液,每天1次,连续45 d。实验结束后测定各组大鼠血铅含量及血清中总抗氧化能力(T-AOC)、丙二醛(MDA)含量、超氧化物歧化酶(SOD)活力及抑制羟自由基能力。结果染铅组大鼠血铅含量和血清中MDA含量高于对照组,血清中T-AOC、SOD活力、抑制羟自由基能力低于对照组,差异有统计学意义(P<0.05)。各剂量TZC拮抗组SOD活力、T-AOC和抑制羟自由基能力均介于对照组和染铅组之间;且随着TZC剂量的升高,染铅大鼠血清中MDA含量呈先下降后升高的趋势,血清中T-AOC、SOD活力及抑制羟自由基能力均呈先升高后下降的趋势,并均以50 mg/kg TZC拮抗组最接近对照组。结论牛磺酸锌可拮抗铅导致的大鼠脂质过氧化损伤。Objective The aim of this study was to investigate the antagonistic effects of taurine-zinc(TZC) on lipid peroxidation in rats exposed to lead. Methods Three weeks old healthy male SD rats were randomly divided into five groups, respectively. The groups were the control group, the lead group and treatment group with 10, 50, 100mg/kg TZC, with eight rats in each group. Rats in all except the control group were given lead acetate solution orally(gavage dosage of 30mg/kg) once a day for 45 days. TZC groups were gavaged with the taurine-zinc solution one hour before lead was given(with dosages of 10mg/kg, 50mg/kg, 100mg/kg,respectively). The blood lead, serum total antioxidant capacity(T-AOC), malondialdehyde(MDA) content, superoxide dismutase(SOD) activity and the ability to inhibit hydroxyl free radicals were measured at the end of the experiment. Results Blood lead and serum MDA content for the lead group were significantly higher than those for the control group, while serum T-AOC, SOD activity,ability to inhibit hydroxyl free radicals for the lead group were lower than those for the control group, with statistically significant difference(P〈0.05). T-AOC, SOD activity and the ability to inhibit hydroxyl free radicals for the TZC treatment groups were between those for the control group and the lead group. And with the increasing TZC dosage, MDA content in serum decreased first and then increased. Serum T-AOC, SOD activity and the ability to inhibit hydroxyl free radicals were decreased after a upward trend. The effects from the 50mg/kg TZC treatment group was the most similar to the effects from control group. Conclusion Taurine-zinc was able to reduce the blood lead content in lead-exposed rats, enhance T-AOC content and SOD activity, increase the ability to inhibit hydroxyl free radicals and reduce the content of MDA. Taurine-zinc had significant antagonistic effects on lipid peroxidation in lead-exposed rats.
