摘  要:目的建立UPLC法同时测定地榆乙酸乙酯部位中没食子酸、儿茶素、没食子儿茶素没食子酸酯和鞣花酸4种鞣质类成分。方法地榆70%乙醇提取液再经乙酸乙酯萃取,甲醇溶解并定容,分析采用shim-pack XR-ODSIII色谱柱(2.0mm×75mm,1.6μm),以甲醇-0.5%醋酸水溶液为流动相梯度洗脱,体积流量0.4mL/min,柱温30℃,PDA检测器于274nm波长下对各成分进行定量测定。结果没食子酸、儿茶素、没食子儿茶素没食子酸酯和鞣花酸在18min内均达到基线分离,各成分在其线性范围内均呈现良好的线性关系,线性范围分别为0.048-0.287μg(r^2=0.999 6),0.151-0.905μg(r^2=0.999 7),0.169-1.014μg(r^2=0.999 4),0.040-0.239μg(r^2=0.999 4)。平均加样回收率分别为98.5%(RSD 1.3%),99.0%(RSD 1.5%),101.2%(RSD 2.1%),99.6%(RSD 1.3%)。结论分析表明内蒙古所产地榆中没食子酸和鞣花酸这两类可水解鞣质的含有量大于其他地区。吉林所产地榆中儿茶素类缩合鞣质的含有量较高。AIM To develop an UPLC method for simultaneously determining four tannins( gallic acid,catechin,gallocatecholgallate and ellagic acid) of ethyl acetate extract from Sanguisorba officinalis.METHODS The crude extract of S.Officinalis was gained through 70% ethanol and ethyl acetate in turn,and then was solved in methyl solution and the volume was set with methanol.Their analysis was performed on shim-pack XR-ODSIII column( 2.0mm × 75mm,1.6μm) with methanol( A) and 0.5% glacial acetic acid( B) as mobile phase in a gradient elution manner.The flow rate was 0.4 m L / min,the detection wavelength was set at 274nm,the injection volume was 2μL,and the column temperature was 30 ℃.RESULTS The peaks of gallic acid,catechin,gallocatecholgallate,and ellagic acid reached the baseline separation within 18 minutes,and showed a good linearity in the range of 0.048 - 0.287μg( r^2= 0.999 6),0.151 - 0.905μg( r^2= 0.999 7),0.169 - 1.014μg( r^2=0.999 4),and 0.040 - 0.239μg( r^2= 0.999 4),respectively.The average recoveries were 98.5%( RSD1.3%),99.0%( RSD 1.5%),101.2%( RSD 2.1%),and 99.6%( RSD 1.3%),respectively.CONCLUSION The experimental data shows that the contents of two constituents of S.officinalis,gallic acid and ellagic acid,collected from Inner Mongolia are higher than these from the rest,the contents of catechin and gallocatecholgallate collected from Jilin Province are the most in the measured samples.
