JC病毒大T抗原及β-catenin在结直肠腺癌中的表达

摘　　要：目的研究JC病毒(JCV)基因编码产物大T抗原(large tumor antigen,T-Ag)的表达及β-catenin(β-连接素)蛋白表达与结直肠腺癌发生发展的关系。方法采用RT-PCR法检测65例结直肠腺癌和20例非肿瘤结直肠黏膜中JCV T-Ag、β-catenin蛋白的表达,并分析其与临床特性及生物学特性之间的相关性。结果在结直肠腺癌中JCV T-Ag和β-catenin阳性表达较正常组织明显升高(P<0.05)。在癌组织中,JCV的表达与组织分化程度及淋巴结转移无明显的相关。β-catenin的表达与组织分化程度及淋巴结转移有明显相关(P<0.05)。结论 JCV T-Ag早期基因编码产物大T抗原在结直肠腺癌的发生、发展中可能起重要作用。β-catenin可以反应肿瘤的侵袭性,为评估肿瘤的预后提供参考。Objective To explore the role of JC virus early region gene encoding product large tumor antigen（T-Ag）andβ-catenin proteins expressions during the colorectal adenocarcinoma occurrence and development.Methods We determined the DNA of JCV and RNA ofβ-catenin in 65 human colorectal adenocarcinoma tissue specimens and 20 cases normal colorectal mucosa,and analyzed the relationship between the expression and biologic characteristic,pathological factors in clinic.Results The JCV T-Ag and positive expression ofβ-catenin in colorectal adenocarcinoma compared with normal tissue were significantly higher（P〈0.05）.Of the JCV expression and tissue differentiation and lymph node metastasis in cancer tissue,there is no significant correlation.Theβ-catenin expression and tissue differentiation and lymph node metastasis was significantly associated（P〈0.05）.Conclusion The JCV T-Ag early region gene encoding product large tumor antigen proteins were over-expressed in colorectal adenocarcinoma and the expressions had relevance to the carcinogenesis and the progression of colorectal adenocarcinoma.Theβ-catenin might play an ortant role in the occurrence,progression and prognosis of colorectal caner.
