摘  要:探讨Pirh2、p53在不同转移潜能乳腺癌细胞系中的表达及其与乳腺癌发生发展的相关性。应用实时荧光定量PCR及蛋白质印迹法检测二者在MCF-10A、MCF-7、MDA-MB-231、MDA-MB-435s细胞株中的表达。Pirh2、p53在正常乳腺上皮及乳腺癌细胞系中表达差异有统计学意义(P<0.05);随着乳腺癌病变程度增加,Pirh2表达水平上升,p53表达水平下降,各组间差异有统计学意义(P<0.05),且Pirh2、p53表达呈显著负相关(P<0.01)。高表达Pirh2可能通过降解p53促进乳腺癌的发生发展。To investigate the expressions of Pirh2 ,p53 in breast cancer cell lines with different invasive ability and its correlation to genesis and development of breast cancer .Real‐time quantitative PCR and Western blot methods were used to detect the expressions of Pirh2 and p53 in MCF‐10 ,MCF‐7 ,MDA‐MB‐231 ,MDA‐MB‐435s cell lines .The expression difference of Pirh2 in breast cancer cell lines and normal breast epithelial cell lines was statistically significant (P〈0 .05);With the increase of invasive ability of breast lesions ,the expression level of Pirh2 was on the rise ,and p53 was on the fall ,the difference was statistically significant in comparison between groups (P〈0 .05) .The expression level of Pirh2 and p53 had significant negative correlation (P〈0 .01) .The high expression of Pirh2 could degrade p53 to promote the genesis and development of breast cancer .
