Effects of Acetic Acid on Expression Levels of Genes Involved in Milk Fatty Acids de Novo Synthesis in Bovine Mammary Epithelial Cells

摘  要:本试验旨在研究不同浓度的乙酸对奶牛乳腺上皮细胞(bovine mammary epithelial cells,BMECs)乳脂肪酸从头合成相关基因表达量的影响。将传至第3代的BMECs悬液接种于细胞培养板上,每孔加入含10%胎牛血清(FBS)的DMEM/F12培养液,于37℃5%CO2培养箱培养48 h。再将培养48 h的奶牛BMECs培养孔随机分配到6组中,每组6个重复,每个重复1个孔。在各组中分别加入含不同浓度乙酸的DMEM/F12培养液,培养液中的FBS用1 g/L无脂肪酸的牛血清白蛋白(BSA)代替,并使反应体系中乙酸的最终浓度分别为0(对照)、4、6、8、10、12 mmol/L。置于37℃5%CO2培养箱继续培养48 h。结果表明:BMECs内甘油三酯的含量随着乙酸浓度的增加呈极显著的一元线性增加(P=0.000),以8-12 mmol/L的乙酸组促进作用较好。脂肪酸合成酶(fatty acid synthase,FASN)和乙酰辅酶A羧化酶(acetyl-coenzyme A carboxylaseα,ACACA)mRNA的相对表达量随着乙酸添加浓度的增加呈极显著的一次线性增加(P=0.000),以8-12 mmol/L乙酸组的表达量较高。硬脂酰辅酶A去饱和酶(stearoyl-CoA desaturase,SCD)mRNA相对表达量随着乙酸浓度的增加,变化趋势不显著(P〉0.05),但从数值上看,所有试验组均高于对照组,以4 mmol/L乙酸组最高。综合得出,乙酸对奶牛BMECs内乳脂肪的合成及其乳脂肪合成相关基因FASN和ACACA mRNA的表达量有极显著的提高效果,其中以培养液中8-12 mmol/L的乙酸效果较好。The purpose of this study was to examine the effects of acetic acid on expression levels of genes in-volved in milk fatty acid de novo synthesis in bovine mammary epithelial cells ( BMECs) . The 3th passage cells were plated in culture plates with DMEM/F12 medium containing 10% fetal bovine serum ( FBS) , then the plates were maintained in an incubator with 5% CO2 and at 37 ℃. After 48 h, cells were distributed to six groups and cultured in DMEM/F12 medium containing different concentrations of acetic acid [0 (control), 4, 6, 8, 10 and 12mmol/L, respectively) , each group consisted of six replicates with 1 pore per replicate. FBS in medium was replaced by 1g/L fatty acid-free bovine serum albumin ( BSA) . Continuous incubation in an in-cubator with 5% CO2 at 37℃ for 48 h. The results showed that triacylglycerol ( TAG) content was significant-ly increased linearly with increasing concentration of acetic acid (P=0.000), and TAG content was greater than that observed in other groups when cells were incubated with acetic acid at 8 to 12 mmol/L. The relative expression levels of mRNA of fatty acid synthase ( FASN) and acetyl-coenzyme A carboxylase α ( ACACA) were significantly upregulated linearly with increasing concentration of acetic acid ( P=0.000 and P=0.000) , especially for 8 and 12 mmol/L. The relative expression levels of mRNA of stearoyl-CoA desaturase ( SCD) was not significantly affected with increasing concentration of acetic acid (P〉0.05), but the expression level in experimental group was higher than that in control group, especially when acetate concentration was 4 mmol/L. Taken together, acetic acid has significant promoting effects on the synthesis of milk fat and ex-pression levels of genes related to milk fat synthesis in bovine mammary epithelial cells, such as FASN and ACACA, and the effects are better when the concentration of acetic acid is 8 to 12 mmol/L.
