摘  要:分析5个不同品种泰山人工北虫草中主要核苷类成分的含量并比较其差异.采用反相高效液相色谱(RP-HPLC)法检测5个样品中尿苷、腺苷、虫草素的含量.色谱柱为KromasilC18(150 mm×4.6 mm,5μm),流动相为甲醇-水(7∶93),流速1.0 mL/min,检测波长260 nm,柱温为室温.3种成分的进样量分别在5.4-540、5.2-520、5.0-500 μg/mL范围与各自峰面积积分值呈良好的线性关系(r分别为0.999 4、0.999 4、0.999 2);平均回收率分别为97.83%、98.10%、98.41%,RSD分别为2.51%、1.46%、2.21%(n均为5).5个品种泰山人工北虫草中主要核苷类成分含量有差异.该方法快速、准确、重现性好,能有效鉴别虫草的核苷类成分,可用于天然虫草和人工虫草中核苷类成分的分析,评价不同来源虫草的质量.To develop a HPLC method for the determination of the nucleosids from cultured Cordyceps, and to study the difference in them. Five samples of Cordycep from different resources were collected. Uridine, adenosine and cordycepin were determined by RP-HPLC method. The separation was performed on a Kromasil C18(150 mm×4.6 mm,5μm) with UV detection at 260 nm, the mobile phase consisted of methanol- water(7 : 93), the flow rate was 1.0 mL/min and the columm temperature was controlled at room temperature. The linear range of uridine, adenosine and cordycepin were 5.4 μg/mL-540 μg/mL (r=0.999 4), 5.2 μg/mL-520μg/mL (r=-0.999 4), 5.0 μg/mL-500 μg/mL (r=0.999 2), respectively. The mean recovery of three components were 97.83 %(RSD=2.51%, n=5), 98.10 %(RSD=1.46 %, n=5), 98.41%(RSD=2.21%, n=5), respectively. The contents of nucleosides in 5 cultured Cordyceps were different from one another. The method is quick, accurate, reproducible and can effectively distinguish nucleosides in various Cordyceps. Consequently it can be used to analyze nucleosides between Cordyceps sinensis and cultured Cordyceps as well as evaluate their quality.
