摘  要:目的探讨大豆皂苷(SS)对环磷酰胺(CTX)化疗荷瘤小鼠疗效及T淋巴细胞的调节作用。方法建立肉瘤S180荷瘤小鼠模型,随机分为模型对照组、CTX组、CTX+SS低、中、高剂量组(10、20、40 mg/kg),连续给药15 d后处死小鼠,测量小鼠瘤重、胸腺和脾脏重量,计算抑瘤率、胸腺指数、脾脏指数;观察小鼠存活时间,计算生命延长率;流式细胞仪测定脾T淋巴细胞亚群百分率。结果化疗后荷瘤小鼠免疫功能低下;SS中、高剂量能增强化疗药物的抑瘤作用,提高化疗小鼠的脾脏指数,延长小鼠的存活时间,提高CD3+、CD4+比例及CD4+/CD8+比值,与CTX组比较差异有统计学意义(P<0.05)。结论大豆皂苷可增强化疗药物的疗效,提高机体免疫力,减轻化疗的毒副作用。Objective To explore the effects of soyasaponins( SS) on the chemotherapy outcome and regulation on T-lymphocyte subsets by cyclophosphamide( CTX) in tumor-bearing mice. Methods The models of S180 tumor-bearing mice were established and the mice were randomly divided into 5 groups: model control group,CTX group and CTX plus SS( 10,20 and 40 mg / kg) groups. All the mice were sacrificed after successive medication for 15 days. The weight of the tumors,spleens and thymuses were measured. The tumor inhibitory rate,the spleen index and the thymus index were calculated. The mice's survival time was observed to evaluate the life extension rate,and the ratios of T-lymphocyte subpopulations in spleen were detected by flow cytometry. Results The immune function of the tumor-bearing mice with CTX chemotherapy was depressed. Middle and high dose( 20 and 40 mg / kg) of SS could enhance tumor-inhibitory effect of CTX,increase the spleen index of the tumor-bearing mice treated with CTX,prolong the survival time and increase the ratios of CD3^+,CD4^+and CD4^+/ CD8^+. There were statistically significant differences between CTX group and middle dose group as well as between CTX group and high dose group( P〈0.05). Conclusions SS can increase the chemotherapeutic efficacy,enhance the immune function and attenuate the toxicity of chemotherapy.
