摘  要:采用平板水解圈法(plate hydrolysis spot method)对分离自酒鬼酒发酵车间空气的细菌进行产酯酶活性菌株筛选,采用基于16S r RNA基因序列的分析方法对高酶活菌株进行系统发育多样性分析。结果表明,73个受试菌株中,有51株酯水解酶活性菌株呈阳性,占受试菌株的69.9%。其中有9株(JSM 2215018、JSM 2215021、JSM2215027、JSM 2215104、JSM 2215109、JSM 2215116、JSM 2215117、JSM 2215126、JSM 2215132)具有较强的酯酶活性。系统发育分析表明,这9株菌属于细菌域(Eubacteria)的4个大的系统发育类群(Actinobacteria,DeinococcusThermus,Firmicutes,Proteobacteria)的8个科(Bacillaceae,Deinococcaceae,Microbacteriaceae,Micrococcaceae,Moraxellaceae,Nocardiaceae,Rhodobacteraceae,Xanthomonadaceae)的8个属:JSM 2215018属于Microbacterium属;JSM 2215021属于Arthrobacter属;JSM 2215027属于Bacillus属;JSM 2215104属于Xanthomonas属;JSM 2215109属于Deinococcus属;JSM 2215116属于Acinetobacter属;JSM 2215117和JSM 2215132属于Paracoccus属;JSM2215126属于Rhodococcus属。研究结果表明,酒鬼酒发酵车间空气源细菌存在较高比例的产酯酶菌株,且这些菌株具有较高的类群多样性。The strains used in this study were isolated from the air samples collected from the fermenting workshop in Jiugui Liquor Distillery by natural precipitation method and culture-dependent method. The esterase-producing capability of 73 strains was investigated by plate hydro-lysis spot method. The results showed that, 51 out of 73 (69.9 %) strains exhibited amylase-producing activity, in which 9 strains (JSM 2215018, JSM 2215021, JSM 2215027, JSM 2215104, JSM 2215109, JSM 2215116, JSM 2215117, JSM 2215126, JSM 2215132) could strongly produce esterase. Then, these 9 strains were investigated by means of phylogenetic analyses based on 16S rRNA gene sequence com-parisons. The results suggested that, these 9 strains were members of 8 genra (Microbacterium, Arthrobacter, Bacillus, Xanthomonas, Deinococ-cus, Acinetobacter, Paracoccus, Rhodococcus) of 8 families (Bacillaceae, Deinococcaceae, Microbacteriaceae, Micrococcaceae, Moraxellace-ae, Nocardiaceae, Rhodobacteraceae, Xanthomonadaceae) in 4 phylogenetic group/phyla (Actinobacteria, Deinococcus-Thermus, Firmicutes, Proteobacteria):JSM 2215018 belonged to the genus Microbacterium, JSM 2215021 Arthrobacter, JSM 2215027 Bacillus, JSM 2215104 Xan-thomonas, JSM 2215109 Deinococcus, JSM 2215116 Acinetobacter, JSM 2215117 and JSM 2215132 Paracoccus, and JSM 2215126 belonged to the genus Rhodococcus. In conclusion, the study results proved that there is abundant bacterial diversity, as well as a number of esterase-pro-ducing strains in the air samples in the fermenting workshop in Jiugui Liquor Distillery.
