摘  要:目的:探讨复方蜥蜴散不同微粒组合剂(简称XY)创新模式(独特配方、不同微粒、糊剂给药)抗癌、阻癌及抑癌的分子生物学机制。方法:将120只SPF级SD大鼠随机抽取20只作为空白组,其余100只采用0.02mol/L的甲基硝基亚硝基胍(MNNG)溶液按5m L/kg灌胃等综合因素造模8周,成功制备胃癌前病变(PLGC)大鼠模型后随机分为模型组、复方蜥蜴散80目组、100目组、80目100目等量混合组(XY80组、XY100组、XY80’100组均为1.5g·kg-1·d-1)、维酶素组(0.3g·kg-1·d-1)共5组,每组13只,均作相应处理后应用免疫组化法检测其对胃黏膜组织P53和Survivin表达的影响。结果:各组大鼠胃黏膜组织P53和Survivin的平均吸光度(A):经统计学分析,模型组与空白组比较,差异有统计学意义(P<0.05);各治疗组与模型组比较,差异有统计学意义(P<0.05);XY各治疗组与维酶素组比较,差异有统计学意义(P<0.05),尤以XY80’100组A值最低,XY80组、XY100组与XY80’100组比较,差异有统计学意义(P<0.05);经Pearson相关分析,Survivin与P53表达呈正相关(P<0.05),相关系数r=0.899。结论:P53和Survivin在PLGC过程中具有协同作用;复方蜥蜴散不同微粒组合剂创新模式不仅配方独特,而且采用80目和100目等量混合和糊剂给药方式可进一步降低凋亡促进基因P53的突变,下调凋亡抑制基因Survivin的表达,从而实现有效干预PLGC和延缓癌变进程的作用。Objective: To explore the molecular biological mechanism of the innovation model (a unique formula, different particles, paste medication way) of Fufang Xiyi powder different combination agent (XY). Methods: 120 rats were randomly divided into control group (n=20) and model establishing group (n=100). Rats in model establishing group received gavage administration with MNNG (5mL/kg) for 8 weeks to establish the precancerous leisions of gastric cancer rat models, then the rats in model establishing group were randomly divided into 5 groups such as model group, 80 mesh of XY group (XY80 group, 1.5g·kg^-1·d^-1), 100 mesh of XY group (XY100 group, 1.5g·kg^-1·d^-1), equal mix of 100 and 80 mesh of XY group (XY80·100 group, 1.5g·kg^-1·d^-1), and vitacoenzyme group (0.3g·kg^-1·d^-1), and with 13 rats in each group. Immunohistochemical method was used to test the expression of P53 and Survivin in gastric mucosa. Results: The average optical density (A) of P53 and Survivin of each group were as follows: the differences between model group and control group, model group and treatment groups, XY groups and vitacoenzyme group were significant (P〈0.05). The A value of XY80·100 group was the lowest, and the differences in A value among XY80 group, XY100 group and XY80·100 group were significant (P〈0.05). The expression of P53 and Survivin is positively related by analyzing of Pearson correlation analysis (P〈0.05), and the correlation index r was equal to 0.899. Conclusion: There is synergistic effect between P53 and Survivin in the process of PLGC. The innovation model of different particles combination agent of XY not only had an exclusive prescription, but also the XY80·100 and paste medication way could reduce mutations of P53 and inhibit the expression of Survivin, in order to interpose PLGC and delay cancerous process.
