摘 要:目的:探讨辣椒素(Cap)对人神经母细胞瘤SH-SY5Y细胞的抑制作用、凋亡率、细胞周期、氧自由基(ROS)、细胞内钙离子(Ca2+)的影响。方法:CCK-8法检测辣椒素(50、100、150、200μmol/L)对人神经母细胞瘤SH-SY5Y细胞的抑制作用;PI染色法检测细胞周期变化;Annexin V-FITC、PI双染检测细胞凋亡;DCFHDA检测ROS水平;Fluo 3-AM检测细胞内Ca2+变化。结果:辣椒素显著抑制SH-SY5Y细胞的增殖,且呈时间和剂量依赖性。辣椒素处理12h后:G2/M期、S期减少,G0/G1期细胞增多(P<0.01),表明辣椒素将SH-SY5Y细胞限定于G0/G1期;早期凋亡率显著增加,为48.35%;辣椒素处理12h后ROS、Ca2+水平显著上调(P<0.01)。结论:辣椒素通过提高细胞内ROS、Ca2+水平,抑制人神经母细胞瘤SH-SY5Y细胞增殖,将其限定于G0/G1期,促进其凋亡,有潜力成为新型抗肿瘤天然药物。Objective: To investigate the effect of capsaicin on inhibition, apoptosis, cell cycle, oxygen radical and Ca2+ of SH-SY5Y human neuroblastoma cells. Methods: CCK-8 method was used to test the inhibition effect of capsaicin with different concentrations (50, 100, 150, 200~mol/L) on SH-SYSY cells, and PI staining method was used to test the cell cycle changes. Apoptosis was detected by using Annexin V-FITC and PI double dye, oxygen radical level was detected by using DCFH- DA method, and Ca2~ concentration was tested by using Fluo 3-AM method. Results: Capsaicin could inhibited the proliferation of SH-SY5Y cell, and in time and dose dependent. Cells in G0/GI phase were increased (P〈0.01), and cells in G2/M and S phase were decreased after 12h treatment of capsaicin, which showed that the capsaicin could block the cell cycle in Gc/Gx phase. After 12h treatment of capsaicin, the early apoptosis rate was 48.35% which increased significantly, and oxygen radical and Ca2+ levels were also increased significantly. Conclusion: Capsalcin could inhibit proliferation of SH-SY5Y cells by increasing oxygen radical and Ca2+ level. It also could block the cell cycle in G0/Gt phase and promote apoptosis of SH-SY5Y cells, so it has the potential to become a new antltumor nature medicine.
