摘 要:目的优选参术活血颗粒中挥发油的提取工艺及β-环糊精(β-CD)包合物的包合工艺。方法采用单因素试验,优选莪术、当归等四味药材的最佳提油工艺;采用L9(34)正交试验设计,以包合率、包合物收得率为考察指标,优选最佳包合工艺。结果挥发油最佳提取工艺为加8倍量水,提取5 h;β-CD最佳包合工艺为:挥发油与β-CD投料比为1∶8(m L∶g),包合时间为60 min,包合温度60℃;应用显微成像法和薄层色谱法(TLC)检验包合物,新的物相中挥发油的成分与包合前基本一致。结论优选出的工艺合理、可行,适合工业化生产。Objective To optimize the extraction process of volatile oil and preparation process of β-cyclodextrin( β-CD) inclusion complex in Shenzhu Huoxue granules. Methods The best oil extraction processes of Curcumae Rhizoma,Angelicae sinensis Radix and other two herbs were optimized with single factor experiment. Using L9( 3^4) orthogonal experiment design,with the inclusion percentage and inclusion complex yield as examining indexes,we screened inclusion process condition. Results The optimal process for volatile oil extraction was adding 8 times of water and extracting five hours. The best inclusion process was as follows: the proportion of volatile oil toβ-CD was 1∶ 8( m L∶ g),mixed for 60 min with an inclusion temperature at 60℃. The components of the new complex were similar to those of the former volatile oil detected by microimaging and thin layer chromatography( TLC). Conclusions The optimized process is reasonable,reliable and suitable for industrialized production.
