Merlin蛋白基因NF2突变与青海回族胃癌易感特点研究

摘　　要：目的研究merlin蛋白基因NF2在青海回族胃癌组织中突变特点。方法通过PCR和DNA测序技术检测merlin蛋白基因NF2外显子的突变情况。结果随机选取5个青海回族胃癌组织中NF2基因外显子,结果检测到第3和第17外显子突变率较高,分别为11.6%、34.8%,突变位点分别位于NF2基因的705、1259处。结论 merlin蛋白基因NF2外显子的突变,可能是导致其失去抑癌功能的原因之一,并与青海回族胃癌易感性有关。Objective To explore the mutation of merlin protein gene NF2 in gastric cancer of Qinghai Hui ethnic group. Methods Using PCR and DNA sequencing to detect the mutation of merlin protein gene NF2. Results The exon 3 and 17 of NF2 showed high mutation level,and the mutation level of exon 3 and 17 were 11. 6% and34. 8%,respectively. The mutation sites were located at 705 and 1259 loci in NF2 gene. Conclusion Exon mutation of merlin protein gene NF2 may be one of the reasons leading to the loss of tumor suppressor function,and it is also related with susceptibility to gastric cancer development of Qinghai Hui ethnic group.
