摘 要:目的研究外源性一氧化氮对正常胃粘膜细胞(GES-1)和两株胃癌细胞(BGC-823,SGC-7901)增殖的影响。方法外源性一氧化氮由硝普钠(Sodium Nitroprusside,SNP)供给,应用MTT法检测一氧化氮对正常胃粘膜细胞和胃癌细胞增殖的影响;应用一氧化氮测试盒检测培养液中一氧化氮浓度。结果与对照组相比,SNP对正常胃粘膜细胞和胃癌细胞的增殖均有抑制作用(P<0.05),随着SNP浓度的增高,培养液中一氧化氮的浓度也随之增高。结论外源性一氧化氮对正常胃粘膜细胞和胃癌细胞的增殖均有抑制作用。Objective To investigate the effect of exogenous nitric oxide on the proliferation of normal gastric mucosa cell line( GES- 1) and gastric cancer cell lines( BGC- 823,and SGC- 7901). Methods Exogenous nitric oxide was produced by the nitric oxide donor sodium nitroprusside( SNP). The inhibitory effects of exogenous nitric oxide on the proliferation of gastric cancer cells were analyzed by MTT assay. Nitric oxide concentration in cell culture medium was detected by standard concentration detection product. Results Compared with the control group,SNP not only inhibited the proliferation of normal gastric mucosa cell GES- 1,but also the gastric cancer cell lines BGC- 823 and SGC- 7901( P 0. 05). Nitric oxide level released from SNP was dependent on its treated concentration. The higher concentration of SNP created the higher level of nitric oxide in cell culture medium was detected.Conclusion Exogenous nitric oxide inhibits the proliferation of both gastricmucosa and gastric cancer cells.
